不同提取溶剂对独一味药材中木犀草素含量的影响

Effects of Different Extraction Solvents on the Content of Luteolin in Lamiophlomis rotate

目的：研究不同提取溶剂对独一味药材中木犀草素含量的影响。方法：采用高效液相色谱法测定木犀草素的含量,分别考察水、50%乙醇、70%乙醇回流提取方法制得的供试品溶液中木犀草素的含量。色谱柱为Diamonsil C18,流动相为甲醇-0.4%磷酸溶液（50∶50,V/V）,检测波长为350 nm,柱温为25℃,进样量为10μl。结果：木犀草素进样量在0.040 8~0.204 0μg范围内与其峰面积积分值呈良好线性关系（r=0.999 8）;精密度、稳定性、重复性试验的RSD≤1.12%;平均加样回收率为98.68%,RSD为0.84%（n=6）;水、50%乙醇、70%乙醇回流提取制备的供试品溶液中木犀草素的含量分别为3.93、4.75、5.93 mg/g。结论：不同提取溶剂制备的供试品溶液中独一味药材中木犀草素的含量不同,从高到低依次为70%乙醇〉50%乙醇〉水。本方法操作简便、结果可靠,可用于独一味药材中木犀草素的含量测定。

OBJECTIVE ： To study the effects of different extraction solvents on the content of luteolin in Lamiophlomis rotate. METHODS ： The content of luteolin in L. rotate was determined by HPLC. The contents of luteolin in test samples prepared by water exlraction, 50% alcohol exlraction and 70% alcohol exlraction were all determined. The determination was performed on Diamonsil C,, column with mobile phase consisted of methanol-0.4%phosphoric acid （50：50, V/V）. The detection wavelength was set at 350 nm and detection temperature was 25 %, the sample size was 10 pl. RESULTS ： The linear range of luteolin were 0.040 8-0.204 0 μg （r = 0.999 8 ） with an average recovery of 98.68 % （RSD = 0.84 %, n = 6）. RSDs of precision, reproducibility and stabiliy tests were lower than 1.12%. The contents of luteolin in test samples prepared by water exlraction, 50% alcohol exlraction and 70% alcohol exlraction were 3.93 mg/g, 4.75 mg/g and 5.93 mg/g, respectively. CONCLUSIONS： The contents of luteolin in test samples prepared by different solvents are different, in descending order as 70% alcohol exlraction〉50% alcohol exlraction〉water exlraction. The method is. simnle and reliable. It can be used for the content determination of L. rotate.
