摘要 目的:对代谢组学分析前慢性肾病患者血浆样品的处理方法进行优化。方法:采用HPLC/TOF-MS技术分别对60例慢性肾病患者及30例健康志愿者的血浆处理方法进行优化研究。结果:实验结果显示,采用甲醇沉淀蛋白质对慢性肾病患者及健康人血浆均可达到较好的除蛋白效果,并且可最大限度地保留血浆中的待测组分;用80%乙腈作为复溶溶剂可最大限度地保留血浆中的待测组分。结论:建立了采用HPLC/TOF-MS技术对代谢组学分析前样品进行处理优化的方法,为研究慢性肾病患者血浆代谢组学奠定基础。 Objective. Patients with chrome kidney disease on plasma sample preparation method metabolomics analysis before opti- mization. Methods. HPLC/TOF-MS technology for 60 cases of patients with chronic kidney disease and 30 healthy volunteers plas- ma research. Results:Experi-mental results show that the protein precipitation with methanol, chronic kidney disease patients and healthy addition to human plasma proteins can achieve better results, and can maximize the retention of analytes in plasma, with 80 aeetonitrile as a solvent reconstitution can be plasma maximize the retention of analytes. Conclusion. the established for metabo- nomics optimization method for sample treatment before analysis using HPLC/TOF-MS technology. To lay the foundation for the study of plasma metabonomics.
