搜风祛痰中药复方对ApoE基因敲除小鼠动脉粥样硬化不稳定斑块HO-1和PPARγ表达的影响 被引量:2

摘要 目的观察搜风祛痰中药复方对Aop E基因敲除小鼠动脉粥样硬化(AS)不稳定斑块血红素氧化酶(HO-1)和过氧化物酶体增殖物激活受体γ(PPARγ)的影响。方法将造模成功的Apo E基因敲除小鼠,分别给予阿托伐他汀以及低剂量、中剂量、高剂量稳斑汤干预,模型对照组采用生理盐水灌胃。给药13周后,取腹主动脉脉,取小鼠主动脉组织HE染色进行病理学观察,并采用免疫组化和RT-PCR技术检测HO-1和PPARγ的表达水平。结果各组小鼠腹主动脉组织中HO-1和PPARγ的表达水平治疗组明显高于模型对照组(P<0.01)。结论搜风祛痰中药复方稳斑汤可能通过影响HO-1和PPARγ的表达而干预AS斑块形成及破裂。 Objective: To observe the effect of Compound Soufengqutanwenban Decoction on unstable plaque in Apo E gene knockout rats with atherosclerosis. Methods: The successful model of the Apo E-gene knockout rats were given low dose,medium-dose and high-dose Soufengqutanwenban Decoction and western medicine for a month. At the same time,the model group was fed by normal saline for one month. After treatment for 13 weeks,aortic tissue was observed under optical microscope pathology after HE staining. And then levels of HO-1 and PPARγ were detected by immunohistochemistry and RT-PCR technique. Results: HO-1 and PPARγ expression levels in aortic tissue of rats in treatment groups were significantly higher than those in control group(P 0.01). Conclusion: Compound Soufengqutanwenban Decoction may stabilize plaques by regulating the expression of inflammatory factors and interfering the formation and rupture of AS plaque,which provides a new idea to prevent AS.
