摘要
目的探讨强心胶囊对心力衰竭大鼠心脏血流动力学及丝裂原活化蛋白激酶(MAPK)信号通路的影响。方法采用盐酸阿霉素腹腔注射法制作心衰大鼠模型,造模成功后的50只大鼠随机分为模型对照组、卡托普利组、强心胶囊低、中、高剂量组。另设正常对照组10只。分别给予生理盐水20m L/kg、卡托普利组100mg/kg、强心胶囊0.2g/kg、强心胶囊0.4g/kg、强心胶囊0.8g/kg、生理盐水20m L/kg灌胃给药4周,4周后观察心衰大鼠血流动力学改变,Western印迹法观察心衰大鼠心肌ERK、P38、JNK的表达情况。结果强心胶囊低、中、高剂量组均可改善心衰大鼠的血液动力学(P<0.05),且对心衰大鼠心率无明显影响,其中强心胶囊高剂量组效果与卡托普利组相仿;与模型对照组比较,强心胶囊中、高剂量组ERK、P38、JNK的蛋白表达水平均不同程度降低,差异具有统计学意义(P<0.05)。结论中、高剂量强心胶囊可改善心衰大鼠血流动力学指标,可能与其通过下调MAPK通路ERK、P38、JNK蛋白的表达有关。
Objective: To investigate the influence of Qiangxin Capsules on hemodynamics of heart failure rats and signal pathway of mitogen -activated protein kinase(MAPK). Methods: Intraperitoneal injection of doxorubicin hydrochloride was used to make heart failure rat model. 50 molded rats were randomly divided into a model group, a captopril group, Qiangxin Capsule groups with low, medium and high dose. 10 normal rats was another control group. Each group was administered orally with normal saline 20mL/kg, captopril lOOmg/ kg, Qiangxin Capsule 0. 2g/kg, Qiangxin Capsule 0. 4g/kg, Qiangxin Capsule 0. 8g/kg, saline 20mL/kg for 4 weeks. The hemodynamics change of the heart failure rats was observed after 4 weeks. Western blotting was used to observe ERK, P38 and JNK expression of the heart failure rats. Results : Qiangxin Capsule groups with low, medium and high dose could improve the hemodynamics of the heart failure rats(P 〈0.05), and had no significant effect on the heart rate of the heart failure rats, in which the effect of the high -dose Qiangxin Capsule group was similar to that of the captopril group. When compared with the model group, the Qiangxin Capsule groups with low and medium dose, the ERK, P38 and JNK protein expression of the high - dose group deduced in different degrees, the difference was statistically significant (P 〈 0.05 ). Conclusion: The medium and high - dose Qiangxin Capsules can improve the hemodynamic parameters of the heart failure rats and may be related to the ERK, P38 and JNK protein expression by down - regulating MAPK pathway.
