摘要
目的研究和分析鞣云实素在不同PH磷酸盐缓冲液中的含量变化情况。方法采用在60℃的恒温条件下观察鞣云实素在不同PH的磷酸盐缓冲液(PH分别为1、2、3、4、5、6、7、8)中的水解情况,应用反相高效液相色谱仪测定鞣云实素及其水解产物没食子酸含量,并比较和分析鞣云实素样品液水解前后鞣云实素和没食子酸含量变化。结果缓冲溶液PH值在1~3范围内,鞣云实素随着PH值的上升,其含量呈逐渐上升的趋势,PH值在4~8范围内,鞣云实素随着PH值的上升,其含量呈逐渐下降的趋势,在PH值1~3范围内没食子酸含量呈逐渐下降的趋势,PH值在4~7的范围内,没食子酸含量呈逐渐上升的趋势,PH值在7~8的范围内,没食子酸含量呈下降的趋势。其中在PH值为1时鞣云实素的水解产物没食子酸含量最高,在PH值为3时没食子酸含量最低;结论鞣云实素在不同PH条件下的含量变化反映了酸碱度对鞣云实素水解反应的影响,本实验研究可为含鞣云实素的中药提取加工、养护、药效学、稳定性研究等方面的应用提供一定参考。
Objective: To study and analyze the content changes of corilagin in different PH phosphate buffers. Methods: The corilagin at a constant temperature of 60℃ was used to observe the hydrolysis of PH 1, 2, 3, 4, 5, 6, 7 and 8 respectively in different PH phosphate buffer, and the corilagin content and their hydrolyzate gallic acid content were determined by RP - HPLC. The content changes of the corilagin and the gallic acid were compared and analyzed before and after the hydrolysis. Results : When PH buffer was in the range of 1 to 3, the corilagin rose with PH value and its content was gradually rising. When PH value was in the range of 4 to 8, the eorilagin rose with PH value, and its content gradually declined. When PH value was in the range of 1 to 3, the gallic acid gradually declined, and when PH value was in the range of 4 to 7, the gallic acid gradually rose. When PH value was in the range of 7 to 8, the gallic acid gradual- ly declined. When the PH value was in the range of 1, the corilagin content and the hydrolyzate gallic acid content were highest. When the PH value was in the range of 3, the gallic acid content was lowest. Conclusion: The content changes of the corilagin in different PH phosphate buffers reflect the influence of Ph value on the corilagin hydrolysis reaction. The study can provide certain reference for the extraction, processing, maintenance, pharmacodynamics and stability study of the cofilagin - containing herbals.
