摘要
目的:探讨恩施地区土家族银屑病患者巨噬细胞移动抑制因子( migration inhibitory factor ,MIF)表达水平,分析M IF173 G/C多态性与银屑病发病关联性。方法对2013年2月至2014年2月恩施中心医院收治的245例银屑病土家族患者及260名土家族健康对照(对照组),应用 SNaPshot SNP 分型技术确定MIF-173 G/C等位基因型, ELISA法检测血清中MIF水平。比较病例组和对照组MIF 表达水平以及MIF-173G/C 基因型和等位基因频率的差异。结果银屑病组MIF水平明显高于对照组( P <0.01); MIF基因173 G /C基因型和等位基因频率在2组间分布差异有统计学意义( P <0.05),银屑病组CC 基因型和C等位基因频率均显著高于对照组( P <0.01);相对于GG基因型,暴露于CG 、CC基因型的相对危险度分别为1.46(95%CI:1.01~2.10, P <0.05)和5.19(95%CI:1.69~15.89, P <0.01);MIF173G/C多态性3种基因型GG、GC、CC的银屑病患者的临床特征比较,显示家族史、寻常型银屑病差异有统计学意义( P <0.05)。结论 MIF-173G/C可能是恩施地区土家族银屑病患者发病的遗传危险因素之一,其机制是该多态性对影响MIF表达水平。
Objective To investigate the levels of macrophage migration inhibitory factor ( MIF) in serum of patients with psoriasis in Tujia population of Enshi District , and to explore the correlation between the pathogenesis of psoriasis and 173 G/C polymorphism .Methods A total of 245 patients with psoriasis ( case group ) who were admitted to our hospital from September 2009 to September 2010 and 260 healthy volunteers of Tujia population in the same period were enrolled in the study.The serum levels of MIF were detected by ELISA , and MIF-173 G/C was genetyped by SNaPshot SNP technique . Finally,MIF-173 G/C genotype and serum levels of MIF were analyzed and compared between the two groups .Results The serum levels of MIF in case group were significantly higher than those in control group [(804.21 ±69.78)pg/ml vs (369.46 ±60.46)pg/ml, P 〈0.01].There were significant differences in the 173G/C genotype of MIF gene and allele frequency between two groups ( P 〈0.05).The CC genotype and C allele frequency in case group were significantly higher than those in control group ( P 〈0.01).As compared with GG genotype,the relative risk exposed to CG,CC genetype was 1.46 (95%CI:1.01-2.10, P 〈0.05),5.19 ( 95%CI:1.69-15.89, P 〈0.01),respectively.The clinical characteristics were compared in the patients with three different kinds of genotype (GG,GC,CC) of MIF173G/C polymorphism,which showed that there was a significant difference in family history ,psoriasis vulgaris ( P 〈0 .05 ) .Conclusion MIF-173 G/C polymorphism may be one of genetic susceptibility factors in pathogenesis of psoriasis in Tujia population of Enshi District , and its potential mechanism is that the polymorphism may affect the expression levels of MIF .
