摘要 目的研究锌指转录因子Slug和血清反应因子(SRF)在食管鳞癌中的表达及其临床意义。方法收集2010年9月至2013年9月食管鳞癌手术根治标本120例,术前均未行放疗、化疗,术后常规取食管鳞癌组织包埋制备蜡块;另取距离肿瘤5 cm的食管组织60例做正常对照。按食管鳞癌诊治规范分析相关资料。所有组织蜡块常规切片三张,一份行常规HE染色,常规病理观察;另两份采用免疫组化法(SABC)检测Slug和SRF蛋白的表达水平,分析其与肿瘤大小、分化程度、浸润深度、淋巴结转移等临床生物学行为的关系。结果 Slug蛋白在食管鳞癌组中的阳性表达率显著高于正常食管黏膜组,差异有统计学意义(P>0.05);SRF蛋白在食管鳞癌组中的阳性表达率显著高于正常食管黏膜组,差异有统计学意义(P<0.05);Slug在食管鳞癌组织中的蛋白表达与肿瘤大小、分化程度、浸润深度以及淋巴结转移差异无统计学意义(P>0.05);SRF在食管鳞癌中的蛋白表达与肿瘤大小、分化程度和是否淋巴结转移无关,差异无统计学意义(P>0.05)。浸润深度中,浆膜+浆膜外组表达阳性率显著高于黏膜+肌层组(P<0.05)。结论 Slug、SRF在食管鳞癌中高表达,并与浸润深度相关,可能参与了食管鳞癌的发生、发展,并在食管鳞癌的侵袭中起到重要的调节作用。 Objective To investigate the expression and clinical significance of zinc finger transcription factor-Slug and serum response factor ( SRF ) in esophageal squamous cell carcinoma ( ESCC ) .Methods One hundred and twenty specimens from ESCC radical surgery without preoperative chemotherapy and radiotherapy were collected from September 2010 to September 2013 .All of ESCC tissues ( ESCC group ) were embedded by paraffin after operation .The other 60 cases of gastric tissues ( control group ) that were 5 cm away from ESCC tissue were served as normal control group .The expression levels of Slug and SRF protein were detected by immunohistochemistry (SABC method).At the same time,the correlations between the expression levels of Slug&SRF and clinical biological behavior of ESCC ,including tumor size ,differentiation degree ,depth of infiltration,lymph node metastasis were analyzed .Results The positive expression rates of Slug and SRF protein in ESCC group were 61.67% (74/120) and 46.67% (56/120),respectively,which were significantly higher than those in control group (13.13%,4/30) and (20.00%,6/30),respectively ( P 〈0.05).There was no significant correlation between the expression levels of Slug protein and tumor size , differentiation degree , depth of infiltration , lymph node metastasis ( P 〉0.05);nor was SRF protein ( P 〉0.05).However the expression levels of Slug and SRF protein of ESCC in serous membrane+serous membrane outside group were significantly higher than those in mucous membrane +muscular layer group ( P 〈0.05).Conclusion The overexpression of Slug and SRF may be related to the depth of tumor infiltration ,which may be involved in the pathogenesis and development of ESCC and may play an important role in the invasion of ESCC .
