重楼皂苷Ⅶ联合顺铂通过内质网应激诱导卵巢癌细胞凋亡 被引量:13

Apoptosis of ovarian cancer cells induced by polyphyllin Ⅶ and cisplatin through endoplasmic reticulum stress

摘要 目的探讨重楼皂苷Ⅶ联合顺铂诱导卵巢癌SKOV3细胞凋亡的分子机制。方法从中草药重楼块根中分离提取重楼皂苷Ⅶ,然后联合顺铂处理SKOV3细胞,分为以下4组:0.1μg/ml重楼皂苷Ⅶ+1μg/ml顺铂组、1μg/ml重楼皂苷Ⅶ+1μg/ml顺铂组、10μg/ml重楼皂苷Ⅶ+1μg/ml顺铂组、对照组。培养24h和48h后,MTT法检测细胞增殖率,Hoechst染色检测细胞凋亡率,Real-time PCR检测凋亡相关基因caspase3以及内质网应激相关基因XBP1和ATF4的表达。结果重楼皂苷Ⅶ联合顺铂能有效促进SKOV3细胞凋亡,其凋亡是通过内质网应激激活caspase途径引起的。结论重楼皂苷Ⅶ能有效抑制肿瘤细胞生长,为临床上治疗卵巢癌提供了实验依据。 Objective To investigate the molecular mechanism of apoptosis of SKOV3 cells induced by polyphyllin VII and cisplatin.Methods The polyphyllin VII was extracted from polyphylla root,then used to treat SKOV3 cells combined with cisplatin.The experiments were divided into four groups:0.1μg/ml polyphyllin VII+1μg/ml cisplatin group,1μg/ml polyphyllin VII+1μg/ml cisplatin group,10μg/ml polyphyllin VII+1μg/ml cisplatin group and con-trol group.24 h and 48 h later,the cell proliferation was detected by MTT,cell apoptosis rate was determined by Ho-echst staining,and the apoptosis-related gene caspase3 and endoplasmic reticulum stress-related gene XBP1 and ATF4 were detected by real-time PCR.Results Combination of polyphyllin VII and cisplatin could be effective in promoting the ap-optosis of SKOV3 ,which was activated through endoplasmic reticulum stress.Conclusion Polyphyllin VII could effectively in-hibit the proliferation of tumor cells,which provided an experimental basis for treatment of ovarian cancer in clinic.
