生长分化因子9(growth differentiation factor 9,GDF9)是第一个被发现的由卵母细胞分泌的生长因子,能直接影响动物的产仔数。G7或c.1111G>A突变是Belclare和Cambridge绵羊GDF9基因编码区1111 bp处发生的G→A突变,导致第371位氨基酸由Val改变为Met(V371M)。G7突变对挪威白绵羊的产羔数有显著影响。Fec GV是高产的Ile de France绵羊GDF9基因编码区943 bp处发生的C→T突变(c.943C>T),导致非保守氨基酸Arg改变为Cys(p.Arg315Cys)。Fec GV突变杂合子绵羊的排卵数和产羔数显著高于野生型。采用测序方法检测Fec GV突变和PCR-RFLP方法检测G7突变在16个山羊品种(济宁青山羊、贵州白山羊、成都麻羊、萨能奶山羊、板角山羊、关中奶山羊、辽宁绒山羊、大足黑山羊、古蔺马羊、波尔山羊、内蒙古绒山羊、金堂黑山羊、川东白山羊、安哥拉山羊、圭山山羊和太行山羊)和6个绵羊品种(小尾寒羊、洼地、湖羊、杜泊、黑萨福克和中国美利奴)中的分布情况。结果表明,在16个山羊品种和6个绵羊品种中均未检测到G7和Fec Gv突变,提示GDF9基因G7和Fec Gv突变对以上山羊和绵羊品种的繁殖力均没有显著影响。
The GDF9 is the first discovered growth factor which was secreted by oocytes and could affect the litter size (LS) of animals. Sequence analyses of the GDF9 coding region showed a single nucleotide polymor- phism (G7 or c. 1111 G 〉 A) in Belclare and Cambridge sheep, responsible for a Val → Met substitution at posi- tion 371 (V371M). The G7 mutation in GDF9 was strongly associated with LS in Norwegian White Sheep. FecGv (c.943C〉T) was identified in prolific Ile de France ewes, resulting in a non-conservative amino acid change (p.Arg315Cys) in the cleavage site of the polypeptide. FecGv heterozygotes ewes had a higher rate of ovulation (OR) and LS than wild-type ewes. The aim of this study was to detect the distribution of the FecGv mutation by re-sequencing. The G7 mutation of the GDF9 gene in sixteen goat breeds (Jining Grey, Guizhou White, Dazu Black, Gulin Ma, Matou, Boer, Chengdu Ma, Saanen Dairy, Inner Mongolia Cashmere, Banjiao, Jintang Black, Guanzhong Dairy, Chuandong White, Angora, Liaoning Cashmere, Guishan and Taihang goats) and six sheep breeds (Small Tail Han, Wadi, Hu, Dorper, Black Suffolk and Chinese Merino sheep) was also detected using the PCR-RFLP method. Results showed that both G7 and FecGv mutations of GDF9 were not detected in all sixteen goat and six sheep breeds tested, indicating that the G7 and FecGv mutations had no significant effect on fecun- dity of these sixteen goat and six sheep breeds.
