血管内皮生长因子在肾间质纤维化大鼠中的表达

摘要 目的分析血管内皮生长因子(VEGF)表达在早期肾纤维化形成过程中的作用。方法 45只雄性SD大鼠右肾切除术后,随机分为模型组(30只)和对照组(15只)。模型组给予腹腔注射牛血清清蛋白,对照组注射生理盐水。分别于第1、5、9周末常规方法测定肾功能、24h尿蛋白(Pr)、VEGF水平,肾脏组织常规病理检测。分析VEGF、Pr、肾小管间质病理(TIL)积分的相关性。结果模型组24h尿蛋白、肾功能和TIL与对照组比较差异均有统计学意义(P<0.05),且出现早期肾间质纤维化的病理特征。第1、5、9周末模型组大鼠VEGF与24hPr水平均呈正相关(P<0.05)。结论血浆VEGF的表达与24hPr水平变化趋势一致,VEGF可能在蛋白尿的发生机制中起重要作用。 Objective To analysis the role of vascular endothelial growth factor (VEGF) in the formation of the early renal fi- brosis. Methods A total of 45 male SD rats after receiving right nephrectomy were randomly divided into model group(30 rats) and control group(15 rats). Rats in the model group were given intraperitoneal injection of bovine serum albumin, rats in the control group were injected with physiological saline. Renal function,24 h urine protein,and VEGF level,routine pathological detection of kidney tissue were detected at first, fifth and ninth week. The correlation between the VEGF level and the renal tubulointerstitial pathological(TIL) score,and urinary protein. Results The 24 h urinary protein, renal function and TIL score in the model group were significant difference compared with those of the control group (P〈0.05), and rats in the model group showed pathological characteristic of the early renal interstitial fibrosis. The level of VEGF and 24 h urine protein excretion in the model group were positively correlated (P〈0.05). Conclusion VEGF might play an important role in the mechanism of proteinuria.
