摘要
目的探讨葛根素对过氧化氢诱导人视网膜色素上皮细胞(ARPE-19 cells)活力损伤和凋亡的影响。方法用200μmol/L过氧化氢(H2O2)诱导氧化应激,不同浓度葛根素作用于ARPE-19细胞24 h,应用WST-1法检测细胞活力,应用流式细胞仪检测细胞凋亡率。结果过氧化氢明显降低人视网膜色素上皮细胞活力,葛根素能剂量依赖性的减少这种降低;过氧化氢明显增加人视网膜色素上皮细胞的凋亡,葛根素能剂量依赖性的减少这种增加。结论葛根素能明显减少过氧化氢诱导的人视网膜色素上皮细胞活力的损伤和细胞凋亡。
Objective To investigate the effect of puerarin on H 2 O2-induced viability loss and apoptosis of ARPE-19 cells.Methods ARPE-19 cells exposed to 200 μmol/L hydrogen per-oxide ( H2 O2 ) were incubated with various concentrations of puerarin for 24 h.Cell viability was detected by WST-1 assay .The cell apoptosis rate was measured by flow cytometry .Results H2 O2 reduced cell viability of ARPE-19 cells.Puerarin reduced H 2 O2-induced cell viability loss in a dose-dependent manner .H2 O2 increased cell apoptosis of ARPE-19 cells.Puerarin re-duced H2 O2-induced cell apoptosis in a dose-dependent manner .Conclusion Puerarin could clearly reduce H2 O2-induced viability loss and apoptosis of ARPE-19 cells.
