姜黄素对大鼠局灶性脑缺血再灌注载脂蛋白E及类视黄醇X受体、肝X受体表达的影响 被引量:5

Effects of curcumin on APOE and RXR、LXR of rats with focal cerebral ischemia-reperfusion injury

摘要 目的:探讨姜黄素(curcumin)对大鼠局灶性脑缺血再灌注模型载脂蛋白E(Apo lipoprotein E,Apo E)及类视黄醇X受体(Retinoic X receptor,RXR)、肝X受体(Liver X receptor,LXR)表达的影响。方法:采用插线法制作大鼠大脑中动脉闭塞局灶性脑缺血再灌注(MCAO)模型。将48只SD大鼠随机分为四组:假手术组(SH);模型组(IR);姜黄素剂量组(100mg/kg、300mg/kg)。再灌注24h,采用盲评法进行神经功能缺损评分估计;测定脑含水量;Western印迹法检测Apo E及LXR、RXR的表达;观察脑组织形态学改变及运用原位细胞凋亡检测(TUNEL法)法检测大鼠海马CA1区神经元凋亡情况。结果:姜黄素在100mg/kg、300mg/kg剂量范围内,可显著改善MCAO大鼠的行为学体征;降低脑含水量;减轻脑组织形态学改变和神经元凋亡;上调APOE及LXR、RXR的表达,姜黄素300mg/kg组治疗效果最为显著。结论:姜黄素具有脑缺血再灌注保护作用,此作用可能与上调APOE及LXR、RXR的表达有关。 Objective: To investigate protective effect and the effect about apolipoprotein E( APOE),retinoic X receptor( RXR),Liver X receptor( LXR) on cerebral ischemia-reperfusion injury in rats. Methods: using Plug-line method make focal cerebral ischemia and reperfusion of middle cerebral artery occlusion model of rat. Adult male SD rats were randomly divided into four groups: sham group( SH); ischemia-reperfusion group( IR); Curcumin-treatment groups: a dose of 100 mg / kg; Curcumin-treatment groups: a dose of 300 mg / kg. Make neurological deficit scores estimate,measure brain water content,Using Western blotting( Western Blot) to detect the expression of APOE,LXR and RXR,Make HE to observe morphological changes of brain tissue,Make TUNEL to detect neurons apoptosis in rat hippocampal CA1 at 24 h. Results: Curcumin in the dose of 100 ~ 300 mg / kg can improve the nerve damage symptoms of MCAO rat; reduce brain water content; relieve brain morphological changes and neuronal apoptosis; increase the expression of APOE,LXR and RXR. Especially,the treatment effect of curcumin 300 mg / kg groups was the most significant. Conclusion: Curcumin can protect cerebral ischemia-reperfusion,This effect may be regulated by increasing the expression of APOE,LXR and RXR.
