摘要
目的:应用益气活血解毒方治疗胶原诱导性关节炎(CIA)大鼠,探讨该方治疗类风湿关节炎(RA)对外周血T淋巴细胞增殖和分化的影响。方法:以牛II型胶原尾根及背部多点注射制作CIA大鼠模型,以免疫抑制剂来氟米特(2mg/kg)为阳性对照药物,益气活血解毒方(5.9g/kg、11.8g/kg、17.7g/kg)干预,流式细胞仪检测大鼠外周血淋巴细胞中T辅助细胞各亚型的比率。结果:大鼠足关节肿胀度观察显示,益气活血解毒方(11.8g/kg、17.7g/kg)可以显著降低CIA大鼠足关节肿胀程度,流式细胞仪检测结果显示,益气活血解毒方可以显著降低CIA大鼠外周血中T辅助(Th)1和Th2的数量和比率,提高Th17细胞比率,效果优于来氟米特组;调节性T细胞(T reg)比率显著升高,益气活血解毒方5.9g/kg、11.8g/kg剂量组与模型对照组相比有显著性差异。结论:益气活血解毒方能够通过调控RA外周血T淋巴细胞的增殖和分化,维持体内自身免疫应答的稳态,抑制炎症反应的发生,缓解因免疫炎症反应造成的关节损伤,这可能是该方治疗RA的关键作用机制。
Objective: To study the proliferation and differentiation of T lymphocytes in collagen II-induced arthritis( CIA) rats treated by Yiqihuoxuejiedu formula. Methods: we used the CIA rats induced by bovine II collagen as animal model and leflunomide as positive medicine control. CIA rats were treated by Yihuoxuejiedu formula( 5. 9g / kg,11. 8g / kg and 17. 7g / kg). The ratios of T lymphocyte subsets in peripheral blood of CIA rats were detected by flow cytometer. Results: The rats’ paw swelling degree was detected by rat paw volume measuring instrument,Yiqihuoxuejiedu formula can significantly reduce the CIA rat paw swelling and has significant difference compared with the model control group( P 〈 0. 01) The ratios of T helper( Th) 1 and Th2 in CIA rats treated by this formula were decreased significantly,the ratios of Th17 were increased observably and T regulated cells in CIA rats had increased markedly which compared to the rats of model group. These effects of Yihuoxuejiedu groups were superior to the leflunomide group. Conclusion: Yihuoxuejiedu formula could regulate the proliferation and differentiation of T lymphocytes in peripheral blood of CIA rats,preserve the balance of autoimmune response and inhibit the inflammation for relieving the joint injury induced by immuno-inflammatory response. It should be the key active mechanism of RA treated by this formula.
