血府逐瘀汤对大鼠肾小球微血管内皮细胞炎性因子MCP-1表达的影响 被引量:5

摘要 目的:观察血府逐淤汤和糖基化终末产物(advanced glycation end products,AGEs)对大鼠肾小球微血管内皮细胞(glomerula endothelial cells,GECs)单核细胞趋化因子(monocyte chemoattractant protein-1,MCP-1)表达的影响,探讨其相关作用机制。方法:对大鼠GECs进行分离培养与鉴定;制备血府逐瘀汤含药血清;采用Western Blot法检测大鼠GECs炎性因子MCP-1蛋白的表达;采用免疫荧光法检测NF-κB活化。结果:与正常对照组比较,施加AGEs刺激因素后MCP-1表达量明显增高,呈剂量(100 mg/L,200 mg/L,400 mg/L)依赖效应;15%血府逐瘀汤含药血清可以抑制AGEs(200 mg/L)的作用,MCP-1的表达量下降;200 mg/L AGEs作用下,大鼠GECs的MCP-1表达量明显增加,成时间依赖效应,同时促进NF-κB活化。结论:AGEs通过促进NF-κB活化,使大鼠GECs的MCP-1蛋白表达量增加,血府逐瘀汤可抑制其作用。 Objective:To investigate the effect of xuefu zhuyu decoction and Advanced glycation end products ( AGEs) on the expression of monocyte chemoattractant protein-1 (MCP-1) in rat glomerular endothelial cells(GECs) and the related mecha-nism. Methods:Primary RGECs were isolated from Sprague-Dawley ( SD) rats. The synthesis of MCP-1 in rat GECs was deter-mined by western blot method. Nuclear factor-kappa B ( NF-κB) activation was detected by fluorescence microscope. Results:AGEs stimulation increased the synthesis of MCP-1 and promoted the activation of NF-κB in rat RMECs. Conclusion:AGEs in-duces MCP-1 production in rat GECs mediated by the activation of NF-κB, which can be inhibited by xuefu zhuyu decoction.
