摘要
目的将贵州小香猪骨髓间充质干细胞(mesenchymalstemcells,MSCs)接种到人羊膜(humanamnioticmembrane,HAM)上,用于治疗放创复合伤全厚皮肤缺损。方法将HAM负载经核荧光标记物DAPI标记的MSCs,培养3~5d后,移植到小香猪全厚皮肤缺损创面作为实验治疗组,以单纯无细胞接种的HAM作对照治疗组。从宏观、光镜、电镜水平进行较系统的观察。结果与对照治疗组相比,HAM负载MSCs能较好地促进放创复合伤皮肤缺损的愈合。荧光标记显示,MSCs参与了局部组织的修复。结论HAM负载MSCs能较好地促进放创复合伤全厚皮肤缺损创面的组织修复。
Objective Bone marrow derived mesenchymal stem cells(MSCs) planted On human amniotic membrane(HAM) were employed to treat full thickness skin wound combined radiation injury in Guizhou minipigs.Methods Cell free HAM and HAM seeded with autologous porcine MSCs labeled with the fluorescent nuclear marker 4′,6 diamidino 2 phenylindole(DAPI) were applied to full thickness skin wound combined radiation injury in Guizhou minipigs.The effects of cell free HAM and HAM seeded with MSCs on this kind of wounds were observed by means of macroscopic observations, light and electron microscopic examinations respectively.Results In contrast to cell free HAM, HAM seeded with MSCs was more valid for facilitating the healing of skin wounds combined radiation injury.MSCs, examined under fluorescent microscope, took part in tissues repair in porcine skin.Conclusion HAM seeded with MSCs was more valid for facilitating the tissues repair of full thickness skin wound combined radiation injury than cell free HAM did in minipigs.
