摘要
以L-酪氨酸、丁醇为主要原料,采用氯化亚砜法和氯化氢法,合成了L-酪氨酸丁酯,产物结构经IR、1H NMR确证。通过单因素试验和正交试验结果表明,氯化亚砜法合成L-酪氨酸丁酯的最佳工艺条件为:物料摩尔比mol(L-酪氨酸)∶mol(氯化亚砜)∶mol(丁醇)=1∶1.3∶10,加热温度120℃,加热时间2 h;氯化氢法合成L-酪氨酸丁酯的最佳工艺条件为:物料摩尔比mol(L-酪氨酸)∶mol(丁醇)=1∶5,通气时间1.5 h,加热温度120℃,加热时间1 h。两种方法均具有原料价廉易得,反应周期短,催化剂易得,产物易分离的特点,适于工业化大规模生产。
L-tyrosine buthyl ester was synthesized with L-tyrosine and butanol as raw materials by the use of thionyl chloride method and hydrogen chloride method. The target compound's structure was confirmed by IR and 1H NMR. Through single factor and orthogonal tests obtained the optimize formulation:material mole ratio (L-tyrosine∶SOCl2∶buthanol) 1∶1.3∶10, reaction temperature 120℃and reaction time 2 h by thionyl chloride method and material mole ratio (L-tyrosine∶buthanol) 1∶5, gas time 1.5 h ,reaction temperature 120℃and reaction time 1 h by hydrogen chloride method. Both methods are suitable for industrial mass production because of cheaply raw materials, short reaction period, readily obtained catalyst and easily separation and purification methods.
