摘要
目的探讨微型RNA122(mi R-122)在细胞水平对乙肝病毒(HBV)复制的影响。方法将mi R-122的表达载体p SU6-mi R-122和HBV感染性克隆p HBV1.3分别转染Hep G2.2.15和Hep G2细胞;酶联免疫吸附试验(ELISA)检测细胞上清中HBs Ag和HBe Ag含量的变化,荧光定量PCR法检测细胞HBV DNA拷贝数的变化。结果mi R-122在Hep G2.2.15细胞和Hep G2细胞中均能够抑制HBs Ag和HBe Ag的表达,还可明显降低HBV DNA的拷贝数。结论mi R-122能够在细胞水平抑制HBV的复制。
Objective To explore the impact of mi R-122 on the replication of hepatitis B virus(HBV) at cell level. Methods Hep G2 and Hep G2.2.15 cells were transfected with mi R-122 and HBV infectious clone p HBV1.3, respectively. HBs Ag and HBe Ag in the supernatants were measured by enzyme linked immunosorbent assay(ELISA). HBV DNA levels were measured by real-time PCR. Results Expression of HBs Ag and HBe Ag levels, as well as HBV DNA copies were down regulated in both Hep G2.2.15 and Hep G2 cells by mi RNA-122. Conclusion mi R-122 inhibits the replication of HBV at cell level.
