摘要 目的通过检测口腔黏膜下纤维性变(oral submucous fibrosis,OSF)组织中GRP78的表达,初步探讨内质网应激在OSF发病中的作用。方法选取OSF患者30例(早、中、晚期各10例)为试验组,健康人5例为对照组,每例取其口腔颊黏膜,免疫组织化学二步法检测2组GRP78的表达情况。结果正常口腔颊黏膜组织中GRP78呈阳性表达,而OSF组织中GRP78表达明显增强,主要表达于上皮层的角质形成细胞,两者的差异具有统计学意义(P<0.05)。OSF早、中期组织中GRP78的表达明显高于OSF晚期组织GRP78的表达(P<0.05)。结论与正常口腔黏膜相比,OSF组织中GRP78的表达明显增高,提示内质网应激可能参与了OSF的纤维化过程。 Objective To investigate the expression and distribution of GRP78 in OSF,and further discuss the possible role of endoplasmic reticulum stress in pathology of OSF. Methods The Envision immunohistochemistry was used for detecting the expression and distribution of GRP78 in tissues of 5 normal controllers and 30 patients suffered from OSF( 10 cases of early,mediate and advanced stage,respectively). Results The expression of GRP78 was mainly detected in keratinocytes. A positive expression of GRP78 was detected in normal oral mucosa,and a significant increasing of expression could be found in OSF( P 〈 0. 05). The expression of GRP78 in OSF was mainly detected in epithelial tissue of keratinocytes cell. Difference existed between early,moderate and advanced stages OSF,while the expression of GRP78 in early and moderate stage were significantly higher than that in advanced stages in epithelium( P 〈 0. 05). Conclusion Comparing with the normal oral mucosa,the expression of GRP78 was increased significantly in OSF tissues. It indicated that endoplasmic reticulum stress may contribute to the pathogenesis of OSF.
