摘要
目的:采用大剂量链脲佐菌素(streptozotocin,STZ)一次性腹腔注射诱导方法建立1型糖尿病大鼠模型,研究性别差异对SD大鼠造模的影响。方法:SD大鼠118只(雌雄各59只),分为雄性对照组(13只),雄性STZ注射组(46只),雌性对照组(13只),雌性STZ注射组(46只)。每只大鼠称重、标记、剪尾取血后用血糖仪测血糖,STZ注射组大鼠一次性腹腔注射60 mg/kg STZ,对照组大鼠一次性腹腔注射等体积枸橼酸盐溶液。分别检测每只大鼠给药后7 d和49 d的空腹血糖值和体重,记录并统计雌性和雄性大鼠成模率及成模后死亡率。结果:与对照组大鼠相比,STZ注射后成模大鼠血糖值显著性升高,雄性与雌性模型大鼠均出现"三多一少"症状。与雌性大鼠相比,雄性大鼠STZ注射后,成模率无显著性差异,而成模后死亡高峰期提前以及死亡率显著性升高。结论:用一次性大剂量STZ诱导SD大鼠建立1型糖尿病模型,雌、雄性大鼠成模率无性别差异,而成模后死亡率出现显著的性别差异。因此在采用SD大鼠进行糖尿病发病机制及其并发症的研究中,应考虑性别因素对研究结果的影响,从而选择双性别。
Objective:To explore the difference of male and female rats in establishing diabetic model by intraperitoneally injecting a high dose streptozotocin( STZ) . Methods:Total 118 male and female SD rats( each 59 rats) were randomly divided into four groups,in-cluding male mode group(n=46),female mode group(n=46),male normal control group(n=13)and female normal control group(n=13). Model groups were induced to type 1 diabetes mellitus by intraperitoneally injecting 60 mg/kg STZ,normal control received in intraperitoneal injection with citric acid buffer. The body weight,fasting blood glucose and the morphology of the spleen ware measured and examined to validate the animal models. Results:Blood glucose of rats was markedly increased after STZ injection. And there were some other symptoms of model rats,such as polyuria,polydipsia and polyphagia,which indicated that type 1 diabetes mellitus had been induced in the models. Compared to male rats,the incidence of female did not changed significantly. Compared to diabetic male rats,the mortality of diabetic female rats decreased significantly. Conclusion:60 mg/kg establishment of male and female rats with type 1 diabe-tes model induced by STZ and there was no significant change of incidence between male rats and female rats. But the mortality of dia-betic rat was changed significantly. Therefore in the pathogenesis of diabetes and diabetic complications should be taken both male rats and female rats into account in the study.
