TLR2及 TLR4在巨噬细胞识别结核分枝杆菌中的作用 被引量:6

摘要 结核病( tuberculosis,TB)是由结核分枝杆菌(Mycobacterium tuberculosis,MTB)引起的一种慢性传染病,主要以呼吸系统感染为主。近年来,结核病日趋严重,位居由单一病原引起患者死亡的严重传染病之首。目前结核分枝杆菌感染者约占全球总人口的1/3,每年约有930多万人发展为活动性结核病,并有200多万人死于该病。结核分枝杆菌感染人体后,主要被巨噬细胞( macrophage)吞噬,未被机体免疫系统清除而潜伏下来的结核分枝杆菌也主要寄生于巨噬细胞内。巨噬细胞是机体防御系统的第一道屏障,一方面发挥着抵抗结核分枝杆菌的作用,另一方面又是结核分枝杆菌体内滞留造成潜伏感染的主要场所。巨噬细胞与结核分枝杆菌的相互作用对结核病产生较大的影响,因此探讨两者的相互作用机制对结核病的防治尤为重要。TLR2、TLR4主要表达于巨噬细胞,并以识别病原相关分子模式( pathogen-associated molecular patterns, PAMPs)为基础,感知病原微生物存在。然后,通过胞内信号传递,或者直接触发胞内杀菌机制,或诱生免疫炎性因子从而扩大非特异性防御作用,或提供共刺激信号,诱导特异性免疫,一般兼而有之。因此,TLR2、TLR4是极为重要的固有免疫受体。
