糖尿病大鼠肾脏8-羟基脱氧鸟苷和超氧化物歧化酶表达的变化及西格列汀对其影响的研究 被引量:9

The expression changes of 8-OHdG and SOD in diabetic rats kidney and their effect of sitagliptin

摘要 目的探讨西格列汀对糖尿病大鼠肾脏8-羟基脱氧鸟苷(8-OHdG)和血清超氧化物歧化酶(SOD)表达的影响。方法复制糖尿病大鼠模型,随机分为西格列汀1mg/(kg·d)干预组(Sita1,n=15),西格列汀10mg/(kg·d)干预组(Sita2,n=15),糖尿病未干预组(DM,n=16)及正常对照组(NC,n=15)。14周末观察各组FPG、FIns、肾功能、SOD 24hUAlb、尿8-OHdG的变化。结果与NC组比较,DM、Sita1和Sita2组FIns及血清SOD活性均下降(P<0.05),FPG、BUN、血肌酐(Scr)、24hUAlb及尿8-OHdG均升高(P<0.05)。与DM组比较,Sita1组BUN、Scr、24hUAlb及尿8-OHdG均降低(P<0.05),血清SOD活性升高(P<0.05);与DM组比较,Sita2组FPG、BUN、Scr、24hUAlb及24h尿8-OHdG降低(P<0.05),FIns和血清SOD活性升高(P<0.05)。结论氧化应激参与糖尿病大鼠肾脏病变的发生发展,西格列汀对糖尿病大鼠肾脏有一定的保护作用。 Objective To investigate the effect of sitagliptin on the expressions of 8-OHdG and SOD in diabetic rats kidney. Methods The rat models of diabetes were randomly divided into the following four groups:Sita1group[Sitagliptin with the dose of 1mg/(kg·d)body weight once daily,n=15],Sita2group[Sitagliptin with the dose of 10mg/(kg·d)body weight once daily,n=15],DM group(untreated diabetes group,n=16),and the NC group(n=15).14 weeks later,the levels of FPG,FIns,BUN,Scr,urinary protein,urinary 8-OHdG and SOD were detected. Results Compared with NC group,FIns and serum SOD significantly decreased in DM,Sita1 and Sita2groups(P〈0.05),while FPG,BUN,Scr,24 hUAlb and urinary 8-OHdG excretion significantly increased in these groups(P〈0.05).Compared with DM group,BUN,Scr,24 hUAlb and urinary 8-OHdG excretion significantly decreased in Sita1group(P〈0.05),while serum SOD significantly increase in this group(P〈0.05);FPG,BUN,Scr,24 hUAlb and urinary 8-OHdG excretion significantly decreased in Sita2group(P〈0.05),in which FIns and SOD significantly increased(P 〈0.05). Conclusion Oxidative stress may take part in the pathogenesis of diabetic kidney injury,and sitagliptin has some renoprotective effect on diabetic rats.
