摘要
目的建立急性中毒患者血浆中百草枯浓度的测定方法。方法采用Hypersil ODS柱(250mm×4.6mm,5μm),流动相为0.1mol·L^-1磷酸液(含75mmol·L^-1庚烷磺酸钠)-乙腈(85:15,三乙胺调pH3),流速1.0mL·min^-1,检测波长258nm,柱温30℃。结果0.02~4.00μg·mL^-1百草枯的线性关系良好(r=0.9999),最低定量浓度为0.02μg·mL^-1,提取回收率为95.48%±3.55%,方法回收率为98.32%~105.16%,日内、日间的RSD均〈10%。共测定42例患者中百草枯的血药浓度169次。结论所用方法快速、准确、灵敏、专一性好,适用于急性中毒患者血浆中百草枯的测定。
OBJECTIVE To establish a sensitive method for the determination of Paraquat in human plasma. METHODS The analysis was conducted on a Hypersil ODS column ( 250 mm× 4.6 mm, 5μm) with the mobile phase consisting of 0.1 mol·L^-1 phosphate(75 mmol·L^-1 sodium beptanesulfonate) -acetonitrile( 85 : 15 ). pH was adjusted to 3 by triethylamine, and the a flow rate of 1.0 mL·min^-1. The detective wavelength was 258 nm and the column temperature was 30 ℃. RESULTS A good linearity was obtained over the range of 0.02 - 4.00 μg·mL^-11 Paraquat ( r = 0. 9999 ) and the detection limit was 0.02 μg·mL^-1. The extraction recovery ratio was 95.48% ±3.55%, and the method recovery ratio was 98.32% - 105.16%. The inter - day and intra - day RSD were both less than 10%. The determinations of 42 patients' plasma concentrations of Paraquat were 169 times. CONCLUSION The method is rapid,accurate, sensitive and specific, which is suitable for the determination of plasma concentration of Paraquat in acute Doisoninz patients.
