健脾解毒中药对Lewis肺癌小鼠脾脏CD4^+CD25^+Treg细胞免疫调节的影响

Effect of Jianpi Jiedu Recipe on Immunological Regulation of Spleen CD4^+CD25^+Treg Cells of Lewis Lung Carcinoma-bearing Mice

目的:研究健脾解毒中药方对Lewis肺癌小鼠脾脏淋巴细胞增殖及CD4+CD25+Treg细胞的影响,探讨健脾解毒中药治疗肺癌的机理。方法:将18只C57BL/6纯系小鼠右腋皮下接种Lewis肺癌细胞0.2ml,随机分为中药组和模型组,并设6只正常鼠对照观察。造模后第5天开始给药,中药组灌服健脾解毒方0.4ml/只,模型组予生理盐水。第14天处死小鼠,CCK-8法检测脾淋巴细胞增殖,流式细胞术检测小鼠脾脏CD4+CD25+Treg细胞含量。结果:荷瘤中药组和荷瘤模型组与正常组比较,脾淋巴细胞增殖功能均有降低,三组差别有统计学意义(P<0.05);荷瘤中药组和荷瘤模型组脾细胞中CD4+CD25+Treg细胞占CD4+T细胞的比例明显高于正常组,三组差别有统计学意义(P<0.05)。结论:实验证实,Lewis肺癌荷瘤环境下,脾细胞的CD4+CD25+Treg细胞的数量增多,脾淋巴细胞增殖低下。健脾解毒中药可增强荷瘤鼠细胞免疫功能。

Objective： To study the effect of TCM on immune function of Lewis lung carcinoma-bearing mice （C57BL/6）. Methods： C57BL inbred mice implanted Lewis lung carcinoma were used model, to observed the influence of Jianpi Jiedu Recipe on tumor growth and the host＇s immune responses. Results： The animal experimental results showed that tumor treatment group and tumor model group and normal group, lymphocyte proliferation had decreased, there was significant difference between the three groups （P〈0.05）. Tumor model treatment group and tumor cells in spleen CD4^＋CD25^＋Treg cells in the ratio of CD4^＋T cells was significantly higher than the normal group, statistically significant differences between the three groups （P〈O. 05 ）. Conclusion： Animal experiments further confirmed, Lewis lung tumor environment, the spleen cells of CD4^＋CD25^＋Treg cells in- crease in the number, the lower lymphocyte proliferation. Jianpi Jiedu Recipe can improve immune function of the tumor-bearing mice.
