摘要
目的:通过观察抗疏健骨颗粒对去势大鼠骨组织PPAγR2mRNA及蛋白表达水平的影响,为抗疏健骨颗粒应用于临床提供分子生物学依据。方法:采用随机区组设计,将50只4~5月龄的雌性SD大鼠分配至假手术组、模型组、小剂量、中剂量、大剂量抗疏健骨颗粒组,给药至12周后检测并分析骨组织PPAγR2mRNA及蛋白表达水平。结果:小剂量组、中剂量组、大剂量组、假手术组骨组织的PPARγ2mRNA灰度值均明显低于模型组(P〈0.05);而小剂量组、中剂量组、大剂量组骨组织的PPARγ2表达水平均明显低于假手术组(P〈0.05);中剂量组骨组织的PPARγ2表达水平均明显低于小剂量组、大剂量组(P〈0.05)。结论:抗疏健骨颗粒对骨组织PPARγ2表达具有一定的抑制作用,可能与抑制破骨细胞活性,调节BMSCs向脂肪细胞或成骨细胞分化的方向,进而影响骨髓成骨细胞和脂肪细胞的数目变化,对绝经后骨代谢异常具有积极的防治作用。
Objective: To investigate the effect of Kangshu Jiangu Granules on the expression of PPAR γ2mRNA and protein expression levels, to provide the molecular biology basis for Kangshu Jiangu Granules used clinically. Methods: A randomized block design was used to allocate 50SD rats to five groups of a model group, low-dose, medium-dose, high-dose group of Kau- gshu Jiangu Granules and the PPARγ2mRNA and protein expression levels of the bone was compared. Results : The gray values of PPARγ2mRNA of low dose group, middle dose group and high dose group, sham group were significantly lower than that of the model group (P〈0. 05); while PPARγ2expression of the low-dose group, middle dose group and high-dose groups were significantly lower than the sham group (P〈0. 05 ) ; PPARγ2expression of the middle dose group were significantly lower than that of the low dose group and high dose group ( P〈0.05 ). Conclusions : Kangghu Jiangu Granules may inhibit osteoclast activity and adjust the direction of BMSCs into adipocytes or osteoblast differentiation, thereby affecting the number of changes of bone marrow into bone cells and fat cells, can product a positive prevention effect on postmenopausal osteoporosis.
