宫颈癌组织中HPV16E6 mRNA与survivin蛋白表达的关系研究

Study on the relationship between HPV16E6 mRNA expression and survivin protein expression in cervical cancer

目的:探讨宫颈癌组织中人乳头瘤病毒(HPV)16E6 mRNA与survivin蛋白表达的情况及其相互关系。方法:选取宫颈癌患者40例、宫颈上皮瘤变患者90例(Cl N 1、Cl N 2、Cl N 3分别30例)及宫颈炎妇女30例,分别检测上述各类患者宫颈组织中HPV16E6 mRNA与survivin蛋白的表达情况,并进行相关性分析。结果:宫颈癌、宫颈上皮瘤变(Cl N 1、Cl N 2、Cl N 3)患者的HPV16E6 mRNA表达水平显著高于慢性宫颈炎患者,且HPV16E6 mRNA表达水平在宫颈癌>Cl N 3>Cl N 2>Cl N1>慢性宫颈炎患者且组间比较差异均有统计学意义(P<0.05)。不同年龄、临床分期、病理类型的宫颈癌患者宫颈癌组织中的survivin蛋白、HPV16 E6蛋白表达差异无统计学意义(P>0.05),不同病理分化的宫颈癌患者survivin蛋白、HPV16 E6蛋白表达差异有统计学意义(P<0.05),低分化患者的survivin蛋白、HPV16 E6蛋白表达显著高于高分化、中分化患者(P<0.05)。对HPV16E6蛋白及mRNA分别与survivin蛋白进行Spearman相关性,结果显示HPV16E6蛋白与survivin蛋白呈正相关关系(r=0.604,P<0.001),HPV16E6 mRNA与survivin蛋白呈正相关关系(r=0.615,P<0.001)。结论:HPV16E6 mRNA与宫颈病变有一定的关系,HPV16E6 mRNA与survivin蛋白在宫颈癌组织中的表达具有一定的关系。

Objective： To explore human papillomavirus（ HPV） 16E6 mRNA and survivin protein expressions in cervical cancer and the interrelationship. Methods： Forty patients with cervical cancer,ninety patients with cervical intraepithelial neoplasia（ CIN）（ thirty patients with CIN Ⅰ,thirty patients with CIN Ⅱ and thirty patients with CIN Ⅲ） and thirty patients with cervicitis were selected; HPV16E6 mRNA and survivin protein expressions in different groups were detected,the correlation was analyzed. Results： The expression levels of HPV16E6 mRNA in cervical cancer group and CIN group were significantly higher than that in chronic cervicitis group,the expression levels of HPV16E6 mRNA in different cervical lesions from high to low were cervical cancer CIN Ⅲ CIN Ⅱ CIN Ⅰ chronic cervicitis,there was statistically significant difference among different groups（ P〈0. 05）. There was no statistically significant difference in the expression levels of survivin protein and HPV16 E6 protein among the cervical cancer patients with different ages,clinical stages and pathological types（ P〉0. 05）; there were statistically significant differences in the expression levels of survivin protein and HPV16 E6 protein among the cervical cancer patients with different pathological differentiation degrees（ P〈0. 05）; the expression levels of survivin protein and HPV16 E6 protein in poorly differentiated cervical cancer patients were statistically significantly higher than those in highly and moderately differentiated cervical cancer patients（ P〈0. 05）. There was a positive correlation between HPV16E6 protein and survivin protein（ r = 0. 604,P〈0. 001）,HPV16E6 mRNA was positively correlated with survivin protein（ r = 0. 615,P〈0. 001）. Conclusion： HPV16E6 mRNA is correlated with cervical lesions to a certain extent,there is a certain relationship between HPV16E6 mRNA expression and survivin protein expression in cervical cancer.
