尾套法与颈动脉插管法测定转基因高血压小鼠血压的比较 被引量:1

EFFECT OF DIFFERENT METHODS ON DETERMINATION OF BLOOD PRESSURE IN HYPERTENSIVE TRANSGENIC MICE

摘要 目的通过比较无创尾套法与有创颈动脉插管法测定血管紧张素原(AGT)-肾素(REN)双转基因高血压小鼠血压,分析二种血压测量方法测定转基因高血压小鼠血压的效果。方法鼠尾基因组PCR方法筛选9只雄性AGT-REN双转基因高血压小鼠,10月龄时分别采用尾套法与颈动脉插管法测定小鼠的收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)以及脉压(PP),与此同时随机选取9只相同背景的野生(WT)小鼠作为对照,对二种方法测得的各项血压值进行比较。结果对AGT-REN双转基因高血压小鼠血压测定结果显示,尾套法测定SBP、PP值均高于颈动脉插管法测得结果,而测定DBP、MAP值均低于颈动脉法测得结果,差异有统计学意义(均P<0.05);对WT小鼠血压测定结果显示,尾套法测定SBP、PP值高于颈动脉插管法测得结果,而测定DBP值低于颈动脉法测得结果,差异有统计学意义(均P<0.05),而MAP无明显差异;二种方法均显示AGT-REN双转基因小鼠各项血压值高于WT小鼠(P<0.05)。结论尾套法和有创颈动脉插管法均可监测转基因小鼠的高血压状态,但所得结果存在显著差异,应根据不同实验选取不同方法,但同一实验中二种方法不可混合使用。针对转基因高血压鼠更适合使用尾套法测量血压值。 Objective To analyze the effects of two methods on blood pressure measurement by comparing the obtained results by tail- cuff and carotid artery catheter in angiotensinogen (AGT) - renin {REN) double transgenie hypertensive mice. Methods Nine male AGT- REN double transgenic hypertensive mice were identified by PCR of genomie DNA purified from tail biopsy samples. The systolic blood pressure (SBP), diastolic blood pressure (DBP), mean arterial pressure (MAP) and pulse pressure (PP} of the transgenic mice were measured by tailcuff and carotid artery catheter at the age of 10 months, and nine wildtype (WT) mice with the same background were randomly selected as control group. The result was analyzed by T test. Results For AGT- REN double transgenie mice, the value of SBP, PP measured by the tail cuff method were higher significantly than that measured by carotid artery catheter method, and the value of DBP, MAP were lower significantly than that measured by carotid artery catheter method {P% 0.05). For WT mice, the value of SBP' PP measured by the tail cuff method were also significantly higher than that measured by carotid artery catheter method, and the value of DBP were lower significantly than that measured by carotid artery catheter method (P〈0.05). However, the values of MAP measured by two methods were not obvious different; The measured results of two methods showed that the blood pres- sure of AGT- REN double transgenic mice was higher than that of WT mice (P〈0.05). Conclusion As the method of carotid artery catheter can do, tail- cuff measurement can also monitor the blood pressure of hypertensive mice, but the results have significant differences. Different methods can be used in different experiment, tail- cuff measurement is more suitable for transgenic mice, but two methods can not be mixed in the same experiment.
