嗜肺军团菌感染小鼠后血清白介素12水平和巨噬细胞功能的变化

The level of serum interleukin 12 and changes in the function of macrophages of mice after Legionella infection

摘要 目的探讨嗜肺军团菌感染Balb/c小鼠后血清白介素12(IL-12)水平和巨噬细胞功能的变化。方法将36只健康Balb/c小鼠随机分为观察组和对照组各18例,观察组小鼠经腹腔注射嗜肺军团菌菌液0.10 m L,对照组小鼠则经腹腔注射等量的生理盐水,分别于注射后1周和1个月,检测并比较两组小鼠的血清IL-12水平和巨噬细胞功能。结果注射1周后,观察组小鼠的血清IL-12水平为(14.98±2.90)Lg/m L,巨噬细胞的杀伤活性为(34.71±7.28)%,较对照组均有明显降低,组间比较差异均有统计学意义(P<0.05);注射1个月后,观察组小鼠的血清IL-12水平及巨噬细胞杀伤活性均有显著回升,组间比较异均无统计学意义(P>0.05)。结论嗜肺军团菌感染Balb/c小鼠后1周,小鼠的血清白介素12水平及巨噬细胞的功能均有显著下降,且于注射后1个月恢复正常,临床上可将感染后1个月作为相关血清抗体的最佳制备时间。 Objective To investigate the serum interleukin 12 (IL-12) level and changes in the function of macro- phages of Balb/e mice after infected by Legionella. Methods 36 healthy Balb/c mice were randomly divided into observation group and control group (each n = 18 ). The observation group was given intraperitoneal injection 0. 10 mL of Legionella pneumophila suspension, while the control group was given equivalent amount of normal saline. After injection for 1 week and 1 month, the serum IL-12 level and function of macrophages of both groups were detected and compared. Results At 1 week after the injection, the serum IL-12 level of mice in the observation group was ( 14. 98 ±2. 90) Lg/mL, and the maerophage killing activity was (34. 71 ±7.28) %, obviously decreased than that of the control group with statistically significant difference ( P 〈 0. 05 ) ; After one month, the level of serum IL-12 and maerophage killing activity in the observation group significantly rebounded; there was no obvious differences between groups (P 〉 0. 05). Conclusion One week after infection with Legionella pneumophila, the serum IL-12 level and macrophage function of the mice significantly decrease, then return to normal after one month. Therefore, the best time for the preparation of the serum antibody should be at one month after the infection.
