黑面神嫩枝叶治疗小鼠慢性皮炎-湿疹有效部位的筛选 被引量:3

Therapeutic effective polar fraction of tender branch-leaves from Breynia fruticosa on chronic dermatitis-eczema in mice

摘要 目的:筛选黑面神嫩枝叶治疗小鼠慢性皮炎-湿疹的有效部位。方法:采用2,4-二硝基氟苯诱导小鼠慢性皮炎-湿疹模型,将120只小鼠分为正常组、基质对照组、模型组、皮炎平组,氯仿、醋酸乙酯、正丁醇及剩余水层(提取物组均设高、低剂量组),每组各10只。每日外用相应药物2次,连续使用12 d。使用游标卡尺测量并计算左右耳厚度差;电子分析天平称量并计算耳肿胀度及肿胀抑制率;解剖胸腺、脾脏称重并计算相应指数;光学显微镜下观察小鼠耳组织病理学变化。结果:黑面神嫩枝叶水提物醋酸乙酯高剂量组和正丁醇组能有效减轻慢性皮炎-湿疹小鼠耳组织增厚、肿胀,显著抑制免疫器官指数,改善其病理学改变,且显示剂量依赖性。结论:黑面神嫩枝叶水提物醋酸乙酯部位和正丁醇部位对小鼠慢性皮炎-湿疹有良好的治疗作用,为进一步寻找其有效成分打下基础。 OBJECTIVE To screen the therapeutic effective polar fraction on chronic dermatitis-eczema in mice of tender branchleaves from Breynia fruticosa. METHODS One-hundred( of) models with chronic dermatitis-eczema were induced with 2,4-dinitrofluorobenzene on mice. One-hundred and twenty mice were assigned into the nomal group,the substrate matched group,the model group,Piyanping group,the hign and low dose of four tested polar fraction groups on average. All models were treated with corresponding drugs for 12 consecutive days,twice per day. Using vernier caliper to measure and calculate the ear thickness difference; using electronic analytical balance to weigh and calculate the ear swelling degree and the inhibition rate; the anatomy of thymus and spleen to weigh and calculate the corresponding index; lastly the changes of ear histopathology in mice of each group were observed by optical microscope. RESULTS The hign dose of ethyl acetate group and N-butyl alcohol group can effectively relieve the ear thickening、swelling and significantly suppress the immune organindex,and improve its pathology change with a dose-dependent relationship. CONCLUSION The ethyl acetate and n-butanol part have good treatment effect on chronic dermatitis-eczema in mice,that lays the foundation for further search for its effective components.
