摘要
目的探讨α-突触核蛋白诱导的人神经母细胞瘤细胞凋亡的分子机制。方法构建α-Syn寡聚体诱导SHSY5Y细胞凋亡模型,流式细胞技术检测SH-SY5Y细胞凋亡;免疫荧光检测SH-SY5Y细胞内α-Syn阳性包涵体;Pull down检测SH-SY5Y细胞Rac1、Cdc42、RhoA活性。结果与对照组相比,α-Syn寡聚体诱导SH-SY5Y细胞凋亡率显著增加,细胞内α-Syn阳性包涵体数量增加,Rac1、Cdc42、RhoA活性增高。结论α-突触核蛋白诱导的人神经母细胞瘤细胞凋亡与Rho GTPases信号通路的活化相关。
Objective To investigate the mechanism of the apoptosis induced by α-synuclein in SH-SY5Y ceils. Methods SH-SYSY cells apoptosis model induced by α-synuelein oligomers was established. Flow cytometry was performed to analyze the SH SYSY cell apoptosis. Immunofluorescence? stain was performed to detect α-synuclein-positive inclusions in SH-SY5Y cells. Pull clown was performed to detect Rho GTPases signaling pathway activation in SH-SY5Y cells. Results Compared with the control group, the percentage of apoptotic cells, α-synuclein-positive inclusions and Rho GTPases signaling pathway in SH-SYSY ceils activity significantly increased in the α-synuclein oligomers group. Conclusion The apoptosis induced by α-synuclein oligomers in SH-SY5Y cells associate closely with Rho GTPases signaling pathway activation.
