摘要
目的探讨SLE人群和正常对照组外周血淋巴细胞中谷氨酰半胱氨酸连接酶(GCL)活性的差异。方法收集30例活动期的SLE病例与30名正常对照,利用密度梯度离心法提取外周血淋巴细胞,酶法测定外周血淋巴细胞中GCL的活性水平。同时分析合并和未合并狼疮肾炎的SLE组之间GCL活性水平的差异。结果与对照组(383.27±18.68mmol/min/mg蛋白浓度)相比较,SLE组(266.10±13.31mmol/min/mg蛋白浓度)GCL活性水平明显下降(P<0.0001);与未合并狼疮肾炎(LN)的SLE组(303.25±18.47mmol/min/mg蛋白浓度)相比较,合并LN的SLE患者GCL平均活性水平(229.64±17.82mmol/min/mg蛋白浓度)显著减低(P<0.009)。结论 SLE患者GCL活性异常,可能在系统性红斑狼疮组发病和病情进展中起一定作用。
Objective To investigate whether glutamate cysteine ligase (GCL)activity is abnormal in peripheral blood mononuclear cells (PBMC)of systemic lupus erythematosus (SLE)patients.Methods The clinical data of 30 systemic lupus erythematosus patients and 30 normal controls were enrolled.PBMC was separated by density gradient centrifugation using the Ficoll Paque system.GCL activity in PBMC was determined by a enzyme fluorescence assay. Results GCL activity was decreased (P 〈0.0001)in SLE patients (266.10±13.31 mmol/min/mg protein)compared with healthy controls (383.27 ± 18.68 mmol/min/mg protein).Moreover,GCL activity in SLE patients with LN (229.64±17.82 mmol/min/mg protein)was significantly decreased(P 〈 0.009)compared with those SLE patients 〈br〉 without LN (303.25±18.47 mmol/min/mg protein).Conclusion Abnormal GCL activity was found in PBMC of SLE patients.GCL activity may play roles in the pathogenesis of SLE.
