摘要
目的:探讨Ech1表达下调对小鼠肝癌淋巴道转移率的影响,及其与肿瘤相关蛋白Anxa7的相互关系。方法:将Ech1基因表达下调的Hca-F细胞植入小鼠足垫,28天后观察小鼠成瘤及转移情况。在Ech1基因表达下调的Hca-F细胞和对照Hca-F细胞中,通过Western blot检测Anxa7表达变化,同时在Anxa7表达下调Hca-F细胞和对照Hca-F细胞中通过Western blot检测Ech1表达变化。结果:下调Ech1基因的表达,使肿瘤细胞体内淋巴结转移率明显降低。Ech1表达下调后,肿瘤转移相关蛋白Anxa7表达增加;Anxa7表达下调后,Ech1蛋白表达下降。结论:肿瘤细胞Ech1基因表达水平与小鼠肝癌淋巴道转移密切相关,Ech1和Anxa7在Hca-F肿瘤淋巴道转移中可能相互作用。
Objective:To observe the lymphatic metastasis of mouse heptocarcinoma when Echl protein was down - regulated in Hca - F cells, and the expression of Ech 1 protein and the tumor related protein Anxa7 in FAnxa7-down cell and FEchl-down, cell. Methods: FEchl downregulatedcell (1 × 106) were inoculated into the feet pad of 615 mice subcutaneously. On the 28th day of postinoculation, the mice were euthanized and their lymph nodes were isola- ted, weighed, sectioned, and stained with hematoxylin and eosin. The expression of Echl and Anxa7 were observed by Western blot analysis in FAnxa7-down cell and FEchl -dow.cell. Results: Down - regulation of Echl could inhibit the lym- phatic metastasis of Hca - F cells, and increase expression of the tumor related protein Anxa7, while down - regulated Anxa7 could decrease the expression of Echl. Conclusion:Echl is an important factor in the lymphatic metastasis of tumors, Echl and Anxa7 may work together during the lymphatic metastasis of Hca- F cell.
