响应面法提取葡萄皮色素及其稳定性的研究 被引量:4

摘要 实验对葡萄皮色素提取工艺及稳定性进行了优化研究。用单因素实验对提取条件进行筛选,通过最速爬坡实验和中心组合设计实验,优化得到了最佳的提取葡萄皮色素的条件。结果表明:乙醇浓度、液料比、提取时间与葡萄皮色素的提取为非线性关系并且对葡萄皮色素的提取影响极为显著。最佳优化提取条件为:乙醇浓度54%、液料比12∶1、提取时间123min、提取pH 1、提取温度70℃,在此条件下葡萄皮色素的最大提取量为37.5μg/mL。用单因素光、热、双氧水、苯甲酸钠进行试验,得出葡萄皮色素对双氧水不稳定,通过正交试验,进一步对葡萄皮色素的稳定性优化。结果表明:各因素对葡萄皮色素的稳定性影响大小为光>热>防腐剂的量,光为阴暗避光处,热为80℃,防腐剂的量为1.0g/L是最佳稳定贮存条件。结论:葡萄皮色素提取条件模型是拟合度高的模型。 The optimization stability is conducted. The research on the extraction extraction conditions are technology of grape skin pigment and its screened by single factor experiment, the optimum extraction conditions of grape skin pigments are optimized by climbing the steepest and central composite design experiments. The results show that the concentration of ethanol, liquid-material ratio, extraction time and extraction of grape skin pigment is of non-linear relationship and their effects on the extraction of grape skin pigment are extremely significant. The optimal extraction conditions are as follows., ethanol concentration is 54%, liquid-material ratio is 12 : 1, extraction time is 123 min, pH is 1 and extraction temperature is 70 ℃. Under such conditions, the maximum extraction amount of grape skin pigment is 37.5 μg/mL. The test of univariate light, heat, hydrogen peroxide, sodium benzoate for grape skin pigment derives that hydrogen peroxide is unstable, orthogonal experiment is for further optimization of the stability of grape skin pigment. The experimental results show that the effect of all factors on grape skin pigment is light〉heat〉 amount of preservatives in turn, the superior stability level is: in clark place, at 80 ℃, preservatives amount of 1.0 g/L. To sum up, the models of grape skin pigment extraction conditions are fitted with high models.
