摘要
目的建立HPLC法定量测定泰脂安胶囊(女贞叶乙醇提取物)中5种三萜类化学成分。方法泰脂安胶囊甲醇超声提取物的HPLC分析采用大连依利特Kromasil C18液相色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-乙腈-10 mmol/L乙酸铵溶液(53∶27∶20),检测波长为205 nm,体积流量为1.0 m L/min,柱温为15℃。结果 19α-羟基熊果酸、2α-羟基熊果酸、3β-反式对羟基肉桂酰氧基-2α-羟基齐墩果酸、齐墩果酸、熊果酸等5种成分分离良好,分别在0.057~0.57μg、0.099~0.99μg、0.016~0.16μg、0.300~3.00μg、0.691~6.91μg范围内具有良好的线性关系,平均回收率均大于97%(RSD〈1.5%,n=6)。结论 HPLC法克服薄层扫描法难以将齐墩果酸和熊果酸二者有效分离的缺点,操作简便,结果准确,灵敏度高,重复性好,能有效控制泰脂安胶囊的质量。
AIM To establish a method for determining five triterpenes in Taizhian Capsules(ethanolic extract of leaves of Ligustri lucidum Ait) by HPLC.METHODS An HPLC method was developed for the determination using Kromasil C18(4.6 mm × 250 mm,5 μm) column,a mixture of methanol-acetonitrile-10 mmol/L ammonium acetate(53 ∶ 27 ∶ 20) were applied as mobile phase.The detective wavelength was set at 205 nm.The flow rate was 1.0 m L / min and the column temperature was maintained at 30 ℃.RESULTS The five constituents were isolated well.The linear ranges of 19α-hydroxy ursolic acid,2α-hydroxy ursolic acid,3β-trans-pcoumaroyloxy-2α-hydroxy-oleanolic acid,oleanolic acid,and ursolic acid were 0.057-0.57 μg、0.099-0.99μg、0.016-0.16 μg、0.300-3.00 μg,and 0.691-6.91 μg,respectively.Their average recovery was above97%(RSD〈 1.5%,n = 6).CONCLUSION The method is convenient,accurate,reproducible and sensitive,which can overcome a shortage of TLC difficult to separate oleanolic acid and ursolic acid.It can be used to control the quality of Taizhian Capsules.
