摘要 目的建立了银杏磷脂软胶囊中12种黄酮类化合物的超高效液相色谱-四级杆-飞行时间质谱法(UPLC-Q-TOFMS)快速筛查方法。方法银杏磷脂软胶囊甲醇提取物分析以Agilent Poroshell 120 EC-C18(2.1 mm×100 mm,2.7μm)为色谱柱;以乙腈(A)-0.1%甲酸溶液(B)为流动相进行梯度洗脱;体积流量0.2 m L/min;质谱定性采用飞行时间质谱,负离子模式扫描。结果在优化的液质联用条件下,通过对照品、数据库匹配及文献数据,鉴定出银杏磷脂软胶囊中12个黄酮类化合物为(1)3-O-[6-O-(α-L-rhamnosyl)-β-D-glucosyl]-3'-methymyricetin,(2)rhamnosyl-glucosyl methymyricetin,(3)quercetin 3-O-[2-O,6-O bis(α-L-rhamnosyl)-β-D-glucoside],(4)quercetin 3-O-[6-O-(α-L-rhamnosyl)-β-D-glucoside],(5)quercetin 3-O-[2-O-(β-D-quercetin 3-O-[2-O-(β-D-glucosyl)-α-L-rhamnoside],(6)Kaempferol 3-O-[6-O-(α-L-rhamnosyl)-β-D-glucoside],(7)rhamnosyl-glucosyl-apigenin,(8)apigenin7-O-(β-D-glucosyl)(9)槲皮素(quercetin)、(10)芹菜素(apigenin)、(11)山柰酚(kaempferol)及(12)异鼠李素(isorhamnetin)。结论该方法可作为测定银杏磷脂软胶囊总黄酮的手段。 AIM To use ultra performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS) for the rapid test of twelve flavonoid compounds in Yinxing Linzhi Soft Capsules.Methods The separation of Yinxing Linzhi Soft Capsules was performed on Agilent Poroshell 120 EC-C18(2.1 mm × 100 mm,2.7 μm) with a mobile phase of formic acid aqueous solution and acetonitrile.The flow rate was at 0.2 m L / min.TOFMS was applied for qualitative analysis under positive ion mode.Results Under optimized conditions,twelve flavonoid compounds were identified by the standard references,flight time of mass spectrometry and literature data.(1) 3-O-[6-O-(α-L-rhamnosyl)-β-D-glucosyl]-3'-methymyricetin,(2) rhamnosyl-glucosyl methymyricetin,(3) quercetin 3-O-[2-O,6-O bis(α-L-rhamnosyl)-β-D-glucoside ],(4)quercetin 3-O-[6-O-(α-L-rhamnosyl)-β-D-glucoside],(5) quercetin 3-O-[2-O-(β-D-quercetin 3-O-[2-O-(β-D-glucosyl)-α-L-rhamnoside],(6) Kaempferol 3-O-[6-O-(α-L-rhamnosyl)-β-D-glucoside],(7)rhamnosyl-glucosyl-apigenin,(8) apigenin 7-O-(β-D-glucosyl),(9) quercetin,(10) apigenin,(11)kaempferol,and(12) isorhamnetin.CONCLUSION The method is accurate,rapid and sensitive,and it can provide a reliable and effective technique for the rapid test of Yinxing Linzhi Soft Capsules.
