摘要
目的建立HPLC同时测定陕西、河南、山西(太原和沂州地区)、山东和河北省产的远志中6种寡糖酯类成分的方法。方法远志甲醇提取液分析的色谱柱为Kromasil C18(200 mm×4.6 mm,5μm);流动相为乙腈(A)-0.1%甲酸溶液(B),梯度洗脱;检测波长330 nm。结果 3,6'-二芥子酰基蔗糖、西伯利亚远志糖A5、球腺糖A、tenuifoliside A、tenuifoliside B、tenuifoliside C的线性范围依次为0.109~4.36μg、0.044 3~1.77μg、0.012 2~0.488μg、0.055 0~2.20μg、0.030 7~1.23μg、0.014 2~0.568μg,平均加样回收率为97.2%~100.3%。结论 6个不同产地远志饮片中3,6'-二芥子酰基蔗糖、西伯利亚远志糖A5、球腺糖A、tenuifoliside A、tenuifoliside B、tenuifoliside C的含有量分别为0.442%~0.867%、0.090%~0.272%、0.044 1%~0.080 5%、0.216%~0.283%、0.101%~0.124%和0.037 4%~0.081 8%.
AIM To develop a quantitative HPLC method for determining six kinds of oligosaccharide esters from Polygala tenuifolia collected from different growing areas,including Shaanxi,Henan,Shanxi(Taiyuan and Yizhou district),Shandong,Hebei province,China.METHODS The P.Tenuifolia methanol extract was separated by Kromasil C18(200 mm × 4.6 mm,5 μm).The mobile phase of water contained acetonitrile(A) and0.1% formic acid water(B) in a gradient elution manner,detection wavelength was set at 330 nm.RESULTS The linear relationships of 3,6'-di-O-sinapoyl-sucrose、sibricose A5、glomeratose A、tenuifoliside A、tenuifoliside B,and tenuifoliside C were in the ranges of 0.109-4.36 μg,0.044 3-1.77 μg,0.012 2-0.488 μg,0.055 0-2.20 μg,0.030 7-1.23 μg,and 0.0142-0.568 μg,respectvely.Their mean recoveries were97.2%-100.3%.CONCLUSION Their contents of 3,6'-di-O-sinapoyl-sucrose、sibricose A5、glomeratose A、tenuifoliside A、tenuifoliside B,and tenuifoliside C in Polygala tenuifolia pieces from six growing areas are in the ranges of 0.442%-0.867% 、0.090%-0.272% 、0.044 1%-0.080 5% 、0.216%-0.283% 、0.101%-0.124%,and 0.037 4%-0.081 8%,respectively.
