响应面分析法优化北芪菇多糖提取工艺研究 被引量:7

Optimization of Polysaccharides Extraction from Astragalus Mushroom by Response Surface Methodology

摘要 研究北芪菇多糖提取工艺和最佳提取条件,为有效开发利用北芪菇奠定试验方法学基础。主要采用水回流提取多糖,并用苯酚-硫酸法测定北芪菇的多糖含量,对温度、时间和液料比进行单因素试验,根据Box-Benhnken的中心组合试验原理,确定响应面试验方案,做出响应面试验结果。通过Design-Expert软件程序进行分析,从而确定最佳提取条件。液料比是影响北芪菇多糖提取的重要因素。水回流提取多糖最佳工艺条件为:温度90.28℃,时间2.06 h,液料比32.19∶1(m L/g)。在该条件下,北芪菇多糖提取率预测值为6.160 78%,验证值为6.71%。该方法测定多糖含量操作简单、省时、高效,可为今后北芪菇多糖的更深入研究与开发利用奠定理论基础。 The purpose was to study the extraction techniques and the optimum extraction conditions of polysaccharides from Astragalus mushrooms in order to lays an experimental methodological foundation for the efficient development and utilization of Astragalus mushrooms. The content of Astragalus mushroom polysaccharides were determinated by water reflux extraction and phenol-sulfuric acid method. The single factor experiment was carried out on temperature, time, and liquid-solid ratio. Scheme and test results were determined using a Box-Benhnken experimental design. Finally the best extraction conditions were determined through the Design-Expert analysis sottware program. The liquid-solid ratio is the important factors that affect Astragalus mushroom polysaccharides exWaction. The optimum conditions of water reflux extraction is: temperature of 90.28 ℃, time of 2.06 h, liquid-solid ratio of 32.19 : 1 (mL/g). Under such conditions, the predicted and experimental values of Astragalus mushroom polysaccharides extraction yield were 6.160 78% and 6.71%. The method was simple operation, time-saving and efficient. It could provide theoretical foundation for the further research and development ofpolysacchafide in Astragalus mushroom.
