摘要
目的观察麻杏二三汤对哮喘小鼠肺组织气道黏蛋白(MUC)5ac m RNA的影响,探讨其抑制哮喘小鼠气道黏液高分泌的作用机制。方法雌性BALB/c小鼠60只,随机分成空白对照组、哮喘模型组、强的松组及麻杏二三汤高、中、低剂量组,每组10只。除空白对照组,另五组以腹腔注射0.1%卵白蛋白(OVA)与10%氢氧化铝混悬溶液致敏,以1%OVA雾化液雾化攻击制作哮喘小鼠模型。分别给予生理盐水、生理盐水、强的松(10.0mg/kg)、麻杏二三汤高剂量(30g/kg)、中剂量(15g/kg)、低剂量(7.5g/kg)灌胃,R-T PCR测定各组MUC5ac m RNA表达。结果麻杏二三汤高、中、低剂量组MUC5ac m RNA荧光定量△Ct值分别为(12.80±0.84)、(12.15±0.49)、(11.41±1.49),均高于哮喘模型组的(9.82±0.84)(P<0.01)。结论麻杏二三汤能够有效抑制哮喘小鼠气道黏液高分泌,其机制可能与其下调支气管肺组织中MUC5ac m RNA表达有关。
Objective To investigate the influence of Maxing Ersan decoction on the expression of pulmonary mucin MUC5ac in asthmatic mice. Methods Sixty female BALB/c mice were randomly divided into 6 groups:blank control group, asthma model group, prednisone-treating group, and Maxing Ersan decoction groups(high-,middle-, and low-dose), with 10 mice in each group. A fresh mixture of 0.1%ovalbumin(OVA) and 10% aluminum hydroxide suspension solution was intraperitoneally injected to sensitize and 1% OVA atomized liquid was used to induce asthma in 5 groups except blank control group. Mice in the corresponding group were given normal saline, normal saline, prednisone(10.0mg/kg), Maxing Ersan decoction(7.5g/kg), Maxing Ersan decoction(15g/kg),Maxing Ersan decoction(30g/kg) by gavage. Real-time fluorescence quantitative polymerase chain reaction assay(RT-PCR) was used to detect the expression of MUC5ac mRNA in mice’ lung tissue of each group. Results TheΔCt value of all Maxing Ersan decoction groups(12.80±0.84, 12.15±0.49, 11.41±1.49)were higher than that of asthma model group(9.82±0.84, P〈0.01). Conclusion Maxing Ersan decoction can effectively inhibit the asthmatic mice’ airway mucus hypersecretion by downregulating the expression of MUC5ac mRNA.
