摘要
探究了以抗坏血酸(AA)还原氯金酸(HAuCl4)来制备免疫检测用胶体金的工艺条件.以粒径及其分布为主要指标,对制备过程中反应时间、反应温度、溶液pH和n(AA)∶n(HAuCl4)四个影响因素进行了研究,通过Box-Behnken响应面试验优化得到最优制备工艺:n(AA)∶n(HAuCl4)=1∶1,反应时间为2h,温度为20℃,溶液pH=11.该条件下制得胶体金的粒径为(45.73±2.41)nm,PDI=0.207±0.013.该制备条件温和,所得胶体金的粒径分布较窄,可以用于免疫检测.
The synthesis of colloidal gold used in immunoassay via the reduction of HAuCl4 by ascorbic acid(AA)has been studied,with focus on the optimization of preparation technique.The effects of reaction time,temperature,pH and n(AA)∶n(HAuCl4)that influence the size and its distribution of colloidal gold have been studied by optimizing the preparation parameters through Box-Behnken Design RSM.The optimal conditions were found as follows:n(AA)∶n(HAuCl4)=1∶1,pH=11,and reacting under 20 ℃for 2hours.The size of gold nanoparticle we prepared was(45.73±2.41)nm,and PDI was 0.207±0.013.The colloidal gold nanoparticles are synthesized under mild condition and have a narrow size distribution,and can therefore be used for immunoassay.
