XRCC3、XPD基因多态性与职业性慢性铅中毒易感性关系病例-对照研究 被引量:1

Matched case-control study on relationship between polymorphisms of gene XRCC3,XPD and the susceptibility of occupational chronic lead poisoning

摘要 目的探讨X射线交错互补修复基因3(XRCC3)Thr241Met和人类着色性干皮病基因D(XPD)Asp312Asn及Lys751Gin与职业性慢性铅中毒的关系。方法选择103名蓄电池厂确诊为职业性慢性铅中毒的男性工人为病例组,按照工种和年龄1:1匹配选择同厂无诊断为铅中毒的铅作业男性工人为对照组。采集2组人群周围静脉血,采用聚合酶链反应一限制性片段长度多态性技术检测XRCC3 Thr241Met、XPD Asp312Asn和XPD Lys751Gin位点的多态性,采用条件Logistic回归分析分析不同基因型与职业性慢性铅中毒的关系。结果2组人群中XRCC3 Thr241Met、XPD Asp312Asn和XPD Lys751Gln多态位点的基因型分布均符合遗传学Hardy—Weinberg平衡定律(P〉0.05)。2组人群XRCC3 Thr241Met3种基因型总体分布比较,差异有统计学意义(P〈0.05)。2组人群XPD Asp312Asn和XPDLys751Gin3种基因型总体分布分别比较,差异均无统计学意义(P〉0.05)。XRCC3Thr241Met的CT和‘rr基因型均增加铅中毒易感性[校正比值比分别为2.44和3.37,95%可信区间为(1.17~5.09)和(1.09~11.14)],而XPD Asp312Asn和XPD Lys751Gin不同基因型与铅中毒易感性无关(P〉0.05)。结论XRCC3 Thr241Met的CT或TT基因型与职业性慢性铅中毒相关;而XPD Asp312Ash和XPD Lys751Gln多态性与职业性慢性铅中毒易感性无关。 Objective To explore the relationship between polymorphisms of gene XRCC3 Thr241Met and XPD Asp312Asn, Lys751Gln and occupational chronic lead poisoning. Methods Based on 1:1 matched case-control study, 103 couple lead-exposed male workers were selected as the case group (occupational chronic lead poisoning patients ) and the control group (workers without poisoning manifestations ) from storage battery factories, and their peripheral venous blood was collected. Single nucleotide polymorphisms of XRCC3 Thr241 Met, XPD Asp312Asn and XPD Lys751 Gin were determined by the polymerase chain reaction-restriction fragment length polymorphism method. The relationship between XRCC3, XPD gene polymorphisms and occupational chronic lead poisoning was studied by multiple factor logistic regression model. Results The genotype distribution of XRCC3 Thr241 Met, XPD Asp312Ash and XPD Lys751 Gin between the case group and the control group was in accordance with the Hardy-Weinberg equilibrium (P 〉 0.05 ). There were significant differences between the case group and the control group in the distribution of each genotype of XRCC3 Thr241 Met (P 〈 0. 05 ). There was no significant difference between the two groups in XPD Asp312Ash and XPD Lys751 Gln mutation frequencies (P 〉 0.05). The genotype CT and TF of XRCC3 Thr241Met increased the susceptibility of lead poisoning, with the adjusted odds ratios and 95% confidence intervals (CI) being 2.44(95% CI: 1.17-5.09 ) , 3.37 (95% CI: 1.09-11.14). The gene of XPD Asp312Asn and XPD Lys751Gln had no relationship with lead poisoning (P 〉 0. 05). Conclusion The genotype CT and TT of XRCC3 Thr241Met accounted for higher susceptibility to occupational chronic, lead poisoning. The XPD Asp312Asn and XPD Lys751 Gin polymorphisms were not the susceptible factors to occupational chronic lead poisoning.
