有氧运动对糖尿病大鼠心肌PPARα及脂质代谢相关基因表达的影响 被引量:6

摘要 目的研究有氧运动对糖尿病大鼠心肌细胞PPARα及其所调控的脂质代谢相关基因表达的影响。方法通过高糖高脂饮食联合尾静脉注射链脲佐菌素(STZ)构建2型糖尿病大鼠模型,实验大鼠分为正常对照组(C)、糖尿病组(D)、正常运动组(CE)、糖尿病运动组(DE),每组10只,有氧运动以游泳训练的方式进行,持续8周。荧光定量PCR和western blot检测心肌组织PPARαmRNA及蛋白表达水平,荧光定量PCR检测心肌组织CD36、Glut4、CPT-1、LCAD等基因mRNA表达水平。结果正常大鼠经8周有氧运动后,血清甘油三酯和游离脂肪酸、心肌游离脂肪酸显著下降,心肌PPARαmRNA和蛋白表达水平显著上调(P<0.01);心肌CD36、Glut4、CPT-1、LCAD mRNA表达水平显著上调(P<0.01)。糖尿病大鼠经8周有氧运动后,血清甘油三酯和游离脂肪酸、心肌甘油三酯和游离脂肪酸均显著下降,心肌PPARαmRNA和蛋白表达水平显著下调(P<0.01),心肌CD36mRNA表达水平显著下降(P<0.01),心肌Glut4、LCAD mRNA表达水平显著升高(P<0.01)。结论有氧运动可显著降低糖尿病大鼠心肌脂质含量,抑制糖尿病大鼠心肌PPARα表达;有氧运动对正常大鼠和糖尿病大鼠心肌PPARα表达具有不同的调控作用。 Objective To study the effect of aerobic exercise training on the expression of PPARαand genes necessary for lipid metabolism in diabetic cardiac.Method:Diabetes rat model was made by high-carbohydrate and highfat diet and injection of streptozotocin.The animals were divided to 4groups:control group(C),diabetes group(D),control with exercise group(CE)and diabetes with exercise group(DE).The PPARα,CD36,Glut4,CPT-1,LCAD expression was analyzed by real-time RT-PCR and Western blot analysis.Results:There was a significant decrease in serum triglyceride,serum free fatty acid and cardiac free fatty acid in CE group compared with C group rats.The mRNA and protein expression of PPARαand CD36,Glut4,CPT-1,LCAD mRNA in the heart were significant increased in CE group rats compared with C group rats.As for diabetes rats,the mRNA and protein expression of PPARαin the heart was significantly higher compared with the C group rats and was significantly lower in DE group rats compared with D group rats.Exercise training drown-regulated the CD36 mRNA and up-regulated the Glut4、LCAD mRNA expression of diabetes rats.Conclusion:Exercise training reduced lipid content and PPARαexpression of diabetes heart;and there was a difference effect of aerobic exercise on expression of PPARαin cardiac between healthy rats and diabetes rats.
