摘要
目的:研究固本防哮饮对哮喘缓解期小鼠肺组织中转录激活因子和信号转导子1(STAT1)的调控作用。方法:用卵蛋白(OVA)和RSV对BALB/c小鼠反复致敏刺激复制哮喘缓解期小鼠模型(正常组除外),随机分为模型组、预防组、固本防哮饮组和孟鲁司特钠组(每组15只)。观察各组小鼠的一般行为活动和病理学变化特点,用Real-time PCR法测定肺组织STAT1、IL-5 m RNA表达,Western Blot检测肺组织STAT1蛋白表达。结果:模型组小鼠肺组织可见支气管壁水肿、增厚,周围有嗜酸性粒细胞和中性粒细胞浸润。预防组、固本防哮饮组及孟鲁司特钠组小鼠肺脏病理改变显著改善,支气管壁充血水肿减轻,管壁和平滑肌厚度减小,且嗜酸性粒细胞和中性粒细胞浸润也显著减少。模型组肺组织中STAT1、IL-5 m RNA及STAT1蛋白显著高表达(P<0.01),预防组、固本防哮饮组及孟鲁司特钠组均能降低其表达,且孟鲁司特钠组降低肺组织STAT1 m RNA及蛋白表达更明显。结论:固本防哮饮可以降低哮喘缓解期小鼠肺组织IL-5、STAT1 m RNA及STAT1蛋白表达,减轻气道炎症,抑制哮喘发作并能预防哮喘的发作。
Objective: To explore the effect of Guben Fangxiao Decoction on regulation of STAT1 in mice with asthma in catabasis. Methods: The mice model with asthma in catabasis was established by BALB/c mice sensitization induced by OVA and RSV. The mice were randomly divided into normal group, model group, prevention group, Guben Fangxiao Decoction group and montelukast group, with 15 mice in each group. General activities and pathological characteristic were observed. The STAT1 and IL-5 m RNA expressions in lung tissue were determined by Real-time PCR method. And, STAT1 protein expression in lung tissue was determined by Western Blot method. Results: In the mice lung tissue of model group, the edema and incrassation in bronchiolar walls and infiltration of eosinophil granulocyte and neutrophile granulocyte could be seen. In the prevention group, Guben Fangxiao Decoction group and montelukast group, the pathological improved obviously, for examples, the oedema of bronchiolar walls was alleviated, the thickness of bronchiolar walls and smooth muscle was decreased and the infiltration of eosinophil granulocyte and neutrophile granulocyte was obvious declined. The STAT1, IL-5 m RNA expressions and STAT1 protein expression in the control group were higher than those in other groups(P〈0.01). The STAT1, IL-5 m RNA expressions and STAT1 protein expression were decreased in prevention group, Guben Fangxiao Decoction group and montelukast group, especially the STAT1 protein expression in the montelukast group. Conclusion: Guben Fangxiao Decoction could reduce the STAT1, IL-5 m RNA expressions and STAT1 protein expression in asthma catabasis, which may alleviate the inflammation of air passage, and inhibit and prevent asthmatic attack.
