衍生化GC-MS法同时测定南方红豆杉叶中19种脂肪酸的含量 被引量:1

Simultaneous determination of 19 fatty acids in leaves of Taxus Chinensis var. mairei by derivatization GC-MS method

摘要 目的:建立衍生化GC-MS法同时测定不同来源南方红豆杉叶中19种脂肪酸甲酯化衍生物的含量。方法:采用HP-VOC(60m×320μm,1.8μm)弹性石英毛细管柱,程序升温:初温80℃,以10℃/min升至150℃保持5min,以3℃/min升至195℃保持5min,以2℃/min升至230℃保持10min,总计运行59.5min。进样口温度为250℃,分流比10∶1,进样量0.6μL,载气为氮气,流速为10m L/min。结果:19种脂肪酸的响应峰面积与其相应浓度的线性关系良好(r>0.9976)。加样回收率为94.85%-105.43%,RSD均小于4.0%。结论:该方法简便快速、准确,具有良好的重复性和回收率,可用于南方红豆杉叶中19种脂肪酸成分含量测定。不同来源的南方红豆杉叶中脂肪酸含量存在显著差异。 Objective: To establish the content of 19 fatty acids in leaves of Taxus chinensis var. mairei from different habitats by derivatization GC-MS method for the simultaneous determination. Methods: The analysis was carried on HP-VOC capillary column(60m×320μm, 1.8μm). The temperature was programmed with an initial temperature of 80℃, then increasing to 150℃ by 10℃/min, holding it for 5min, and increasing to 195℃ by 3℃/min, holding it for 5min, then increasing to 230℃ by 2℃/min, holding it for 10 min. The total time was 59.5min. The temperature of the injection port and interface was set at 250℃. The split ratio was 10∶1, the injection volume was 0.6μL. Nitrogen was used as the carrier gas, and flow rate was 10 m L/min. Results: Nineteen fatty acids showed good linearity over the tested ranges(r〉0.9976). The average recoveries of the method were between 94.85%-105.43% with RSD〈4.0%. Conclusion: This method is simple, rapid, accurate with good repeatability and recovery. It is available for the simultaneous determination of the content of 19 fatty acids in leaves of Taxus Chinensis var. mairei. There are significant differences between different sources of fatty acids in the leaves of Taxus chinensis var. mairei.
