毛茛总苷中扩血管活性物质实验研究

Experimental Study on the Active Substance of Expanding Vessel From Total Glycosides of Ranunculus Japoniucus

目的:探讨毛茛总苷中总黄酮含量与其扩血管作用关系。方法:采用大鼠离体胸主动脉环张力测定方法,观察毛茛总苷制备过程中不同浓度乙醇洗脱部位对去甲肾上腺素诱导的胸主动脉环收缩的抑制作用;以芦丁为标准品,采用紫外分光光度法测定对应各部位的总黄酮含量;以总黄酮终浓度为横坐标,以各不同部位对抗去甲肾上腺素引起的血管环收缩的抑制率为纵坐标,进行线性回归,分析二者相关性。结果:毛茛总苷不同部位对去甲肾上腺素诱导的主动脉环张力升高均有不同程度的抑制作用(P<0.05);以总黄酮终浓度对毛茛总苷抑制去甲肾上腺素引起血管环收缩的抑制率进行线性回归,相关系数R2=0.919 9,相关性良好。结论:毛茛总苷中总黄酮类成分可能是引起血管扩张的活性成分。

Objective： To study the relation between the total flavonoid content from the total glycosides of ranunculus japoniucus and its function of expanding vessel. Methods： Observed the inhibition effect of different parts of total glycosides of ranunculus japo- niucus on contraction of thoracic aortic induced by norepinephrine （NE） with the method of determining the tension of the thoracic aortic rings in vitro on the test rats. Measured the total flavonoid content in different parts of total glycosides of ranunculus japoniu- cus by UV spectrophotometry ,and the Rutin was used to be the standard. Created the regression curve by using the total flavonoid contents as the abscissa and the inhibition rates of the different parts of total glycosides of ranunculus japoniucus against the vessel loop as the vertical coordinate, and investigated the correlation regression between the above two by Logistic and linear statistics. Resuits. The different alcohol part of total glycosides of ranunculus japoniucus can restrain the increaseing of the thoracic aortic tension on different degrees which induced by norepinephrine （P^0.05）. Investigated the correlation regression between the total flavonoid content and the inhibition rates of total glycosides of ranunculus japoniucus restraining the contraction of the vessel loop induced by norepinephrine （NE）, and the correlation coefficient is 0. 919 9, which shows a good correlation. Conclusion： The flavonoid in total glycosides of ranunculus japoniucus glycosides might be the active components that result in the vasodilatation.
