摘要
采用蒽酮-硫酸法测定南瓜籽多糖含量并对其测定条件进行研究。结果表明最佳条件为:蒽酮浓度2.0 mg/m L,体积分数80%的硫酸,样品溶液与蒽酮-硫酸试剂加入量的体积比为1∶4,加热时间15 min,冷却后放置10 min、625 nm比色。平均加标回收率为97.3%,RSD 3.98%。测得样品中多糖含量为18.14%。结果表明,该方法适用于南瓜籽多糖含量的测定。
Anthrone-sulfuric acid method was used to determine content of polysaccharide in pumpkin seeds and the method conditions were optimized. The experimental results showed that the optimal operating conditions were : 2 mg/mL anthrone, 80 % sulfuric acid, the volume ratio of the sample solution to anthrone-sulfuric acid was 1 : 4, heated in boiling water bath for 15 min, coloration time of 10 min after cooling with city water and the wavelength for measurement was 625 nm. The average recovery rate was 97.3 % with RSD of 3.98 %. The content of polysaccharide in the sample determined by this method was 18.14 %. The result declared that Anthronesulfuric acid method was rapid, accurate, stable and suitable for detecting the polysaccharide content.
