摘要
玫瑰(Rosa rugosa)组织色素较多,富含酚类物质.为了提取适于SRAP分析的高质量的玫瑰DNA,本实验基于CTAB法,在浓度2%和3%CTAB基础上,设计不同浓度的β-巯基乙醇和PVP抗氧化剂进行5种组合,研究了β-巯基乙醇和PVP对玫瑰DNA提取质量的影响.结果表明,在3%CTAB提取液中添加适量的β-巯基乙醇和PVP能获得纯度较高的玫瑰DNA,在组合Ⅲ(3%CTAB、2%β-巯基乙醇+10%PVP)中,DNA电泳谱带清晰,无明显拖尾现象,SRAP分子标记效果较好,DNA的质量和纯度均满足于SRAP技术和DNA指纹图谱研究的要求.
Rosa rugosa is the plant with high phenolic substance and pigment. In order to extract the high quality DNA suitable for SRAP analysis, the modified method was used with 5 kinds of combinations of different concentration of β-mercaptoethanol and PVP and 2% or 3% CTAB. The results showed that it can get the high purity rose DNA from the extracts of the combination of 3% CTAB+2% β-mercaptoethanol+ 10% PVP. Furthermore, the electrophoresis bands of DNA were clear and exhibited no significant tailing phenomenon. The average yield of rose DNA can meet the quality and purity in SRAP technical and DNA fingerprint requirements.
