酵母菌0732-1处理哈密瓜苗后对其抗病性相关酶活的影响

摘要 【目的】通过酵母菌0732-1处理哈密瓜幼苗,对其抗病性相关酶活的影响,验证P.anomola 0732-1具有诱导寄主抗性的生防类型。【方法】利用P.anomola 0732-1的发酵液和哈密瓜细菌性果斑病菌(Aac)喷布哈密瓜幼苗叶片,通过紫外-可见分光光度计分时段连续测定哈密瓜叶片组织内超氧化物歧化酶(SOD)、过氧化酶(POD)、多酚氧化酶(PPO)、苯丙氨酸解氨酶(PAL)的变化。【结果】P.anomola 0732-1发酵液在一定接种时间内能诱导哈密瓜幼苗叶片4种抗性相关酶活的升高,SOD和POD在整个试验过程中一直保持较高的水平。喷布后24 h时,PAL酶活峰值10.72 U/mg,为对照的1.2倍;36 h时,PPO酶活峰值为4 529.20U/g,是对照的1.4倍;60 h时,SOD和POD酶活显著高于Aac和对照,其中P.anomola 0732-1发酵液诱导幼苗叶片POD酶活峰值为432.86 U/g,是对照的1.8倍。在试验中接种Aac能诱导哈密瓜幼苗叶片POD活性升高。【结论】初步证实了P.anomola 0732-1能够诱导哈密瓜幼苗产生抗性,为P.anomola 0732-1在田间防治哈密瓜细菌性果斑病害提供了依据。 [ Objective ] The project aims to verify Hami melon fruit bacterial bloth Base on changes of resistance - related enzymes in host tissue, inducing Hami melon seedling leaf resistance as biocontrol mode of Pichia anomola 0732 - 1. [ Method ] Time dynamic of phenylalanine ammonia lyase ( PAL), polyphenoloxidase (PPO), superoxide dismutase(SOD) and peroxidase (POD) were detected by using ultraviolet specrophotometer after inoculating Aac and fermentation liquid of P. anomola 0732 - 1 into Hami melon seeding leaves, respectively. [ Result]The four kinds of resistance -related enzyme activities were enhanced significantly in a certain period of time by being treated with P. anomola 0732 - 1 fermented liquid in Hami melon seedling leaf, and SOD or POD always maintained at a higher level in the whole test process. The peak value of PAL activity in seedling leaf was 10.72 U/mg, and 1.2 times that of control after spray 24 h ; The PPO activitycould reach 4 529.20 U/g, 1.4 times that of control at 36 h ; SOD and POD enzyme activity were significantly higher than that of Aac and contrast at 60 h, and POD activity peak value was 432.86 U/g, 1.8 times that of control after spraying P. anomola 0732 - 1 fermented liquid. POD activity increase of Hami melon leaf could be induced by incubation with Aac throughout the experiment. [ Conclusion]This work supported the view that inducing resistance - related enzymes by being treated with P. anomola 0732 - 1 could be involved in the mechanism against Hami melon bacterial fruit blotch. Meanwhile, those results contributed to the establishment of effective management for controling BFB in the field.
