脊神经背根切断大鼠关节软骨组织AQP1表达及意义

摘要 目的探讨水通道蛋白质1(AQP1)在脊神经背根切断大鼠股骨下端关节软骨组织中的表达及意义。方法雄性Wistar大鼠30只,实验组切断右侧L3、L4脊神经后根,对照组只切开皮肤及软组织。术后2、6和10周取材,观察两组股骨下端关节软骨的形态学变化,同时检测两组骨组织中AQP1表达的变化。结果实验组关节软骨出现进行性损伤与退变;各时间点骨组织中AQP1表达与对照组比较,差异有显著性(t=5.075~15.587,P〈0.01)。结论脊神经背根切断可引起关节软骨损伤,而AQP1表达上调可能是软骨损伤早期病理特征之一。 Objective To study the expression of AQP1 and its significance in the articular cartilage of distal femur of L3 -- L4 dorsal rhizotomy in rats. Methods Thirty Wistar rats were divided into experimental group and control group. The right dorsal roots of the third and fourth lumbar (La and L4) nerve of the rats in the experimental group were cut off, that in the control group only the skin and soft tissues were cut through. Specimens of right distal femur were taken 2,6 and 10 weeks after operation for observing the morphologic variations in articular cartilage and measuring the expression of AQP1. Results Progressive da- mage and degeneration appeared in articular cartilage of the experimental group, the expressions of AQP1 at each time point was significantly different as compared with the control group (t =5.075-15.587 ,P〈0.01). Conclusion Rhizotomy may cause inju- ry of articular cartilage, and up-regulation of expression of AQP1 is probably one of the early pathologic features of cartilage damage.
