新疆特高发肿瘤资源库中冷冻组织质量鉴定标准的研究

On the quality identify for frozen tissues from the Xinjiang tumor resource bank

目的在新疆特高发肿瘤资源库中建立冷冻组织质量鉴定体系。方法选取肿瘤资源库中食管癌、宫颈癌、肝癌冷冻组织及正常对照组织各10例,利用HE染色技术检测标本取材的准确性,琼脂糖凝胶电泳鉴定DNA质量,琼脂糖凝胶电泳检测RNA质量,Western-blot鉴定蛋白质质量。结果 HE染色观察检测标本肿瘤部分占80%以上,28s/18s为1.8～2.0,蛋白质及DNA质量均完整无降解。结论建立了一个规范化的冷冻组织质量控制体系,可以作为肿瘤资源库标本质量鉴定的常规方法。

Objective To establish a standard protocol for quality control of frozen tissues in the Xinjiang- Tumor Resource Bank. Methods Ten frozen tissues and normal control tissues of cervical cancer esophagus cancer and hepatocellular carcinoma were respectively randomly selected, stained with HE and pathologically checked. Agarose electrophoresis was used to detect the quality of the DNA. Agarose elec- trophoresis used to check the RNA quality. Western blot was used to detect the quality of the protein from these frozen tissues. Results HE staining showed that all the detected samples contained 80% or greater tumor. 28s/18s ratio were respectively 1.8-2.0. The good quality of the protein and DNA from these fro- zen tissues was observed. Conclusion We have established a standard quality control system, which is ap- plicable as a routine method in quality control of frozen tissues from the tumor resource bank.
