免疫性不孕症患者外周血CD4^+CD25^+调节性T细胞、叉头状转录因子3、细胞毒性T淋巴细胞相关抗原4的水平研究

Research on relationship between frequency of circulating CD^4+ CD25^+ Treg, Foxp3, CTLA-4 and immunological infertility

目的探讨外周血CD4+CD25+调节性T细胞(CD4+CD25+Treg)、叉头状转录因子3(Foxp3)、细胞毒性T淋巴细胞相关抗原4(CTLA-4)与免疫性不孕的关系,为免疫性不孕的诊断和治疗提供依据。方法用流式细胞仪检测60例免疫性不孕妇女(研究组)和60例正常妊娠的育龄妇女(对照组)外周血中CD4+CD25+Treg、Foxp3、CTLA-4水平,并对检测结果进行统计学分析。结果研究组外周血中CD4+CD25+Treg、Foxp3、CTLA-4水平值分别为:(3.66±0.59)%、(2.97±0.63)%、(3.17±0.36)%,均低于对照组[(8.17±0.63)%、(7.17±0.53)%、(8.21±0.56)%],两组比较差异均有高度统计学意义(均P<0.01)。结论免疫性不孕症患者外周血中CD4+CD25+Treg、Foxp3、CTLA-4的表达水平比正常妊娠的育龄妇女少,CD4+CD25+Treg、Foxp3、CTLA-4表达的降低可能是免疫性不孕的重要原因。

Objective To investigate the relationship between the frequency of circulating CD4＋CD25＋Treg, Foxp3,CTLA-4 and immunological infertility, and to provide the basis for the diagnosis and treatment of immunological infertility. Methods The levels of the circulating CD4＋CD25＋Treg, Foxp3, CTLA-4 were detected by flow cytometry（FCM）in 60 cases of immunological infertility women（study group） and 60 cases of health gestation age women（control group）,and the results were analyzed with statistics. Results The levels of the CD4＋CD25＋Treg, Foxp3, CTLA-4 in peripheral blood of the study group were（3.66±0.59）%,（2.97±0.63）%,（3.17±0.36）%, which were lower than those of the control group [（8.17±0.63）%,（7.17±0.53）%,（8.21±0.56）%], the differences between the two groups had statistical significances（all P 〈0.01）. Conclusion The levels of the CD4＋CD25＋Treg, Foxp3, CTLA-4 in immunological infertility women have significantly decreased, which suggestes that the reduce of the gestation CD4＋CD25＋Treg, Foxp3, CTLA-4 may play an important role in female immunological infertility.
