摘要
应用SRAP技术开发了龙眼指纹检索系统并进行了遗传多样性分析。12对引物组合在16份龙眼及龙荔种质中共扩增出257条DNA带,其中248条为多态带(占96.5%),平均每个引物扩增多态性带20.7条。17份材料间的遗传相似系数范围为0.399~0.871。利用Me6Em7、Me5Em4、Me2Em8等3对引物开发的指纹检索系统,可以区分全部17份种质。根据相似系数进行聚类分析,16个龙眼品种和龙荔分成2大组,龙眼品种间的聚类结果基本反映了供试材料之间的亲缘关系。
SRAP was assessed to develope a fingerprinting key and study the genetic diversity on Longan. A total of 257 DNA bands were amplified by 12 selective primers, 248 of which (96.5%) were polymorphic. The average number of polymorphie DNA bands amplified by each primer was 20.7. The genetic similarlity coefficients of 17 materials ranged from 0.399 to 0.871. A fingerprinting key developed by 3 pairs of primers: Me6Em7, MeSEm4 and Me2Em8 could discriminate 17 materials. UPGMA cluster showed that 17 materials were classified into 2 cluster groups: Dimocarpus longana and Dimocarpus confinis, and the result revealed the closed relationships among cultivars tested.
